Chin J Biotech, 2016, 32(1): 摘 要 : 目 前 常 用 的 基 因 修 饰 方 法 是 在 同 源 重 组 介 导 下, 电 转 线 性 片 段 替 换 染 色 体 上 指 定 序 列 因 过 程 错 误 掺 入, 该 方 法 常 常 会 在 同 源 序 列 部 位 产 生 一 些 突 变 为 了 避 免 此 类 突 变, 我 们 建 立 了 一 种 新 的 无 痕 删 除 方 法 首 先 将 含 有 抗 性 标 记 ( 两 侧 带 有 I-Sec I 识 别 位 点 ) 的 线 性 DNA 电 转 到 重 组 感 受 态 细 胞 内, 用 抗 性 基 因 替 换 基 因 组 上 指 定 序 列 ; 然 后, 将 携 带 融 合 同 源 臂 ( 两 侧 带 有 I-Sec I 位 点 ) 的 供 体 质 粒 导 入 上 述 细 胞, 诱 导 表 达 I-Sec I 内 切 酶 切 割 供 体 质 粒 释 放 同 源 片 段, 同 时 切 除 染 色 体 上 抗 性 基 因 产 生 双 链 断 裂, 通 过 分 子 间 同 源 重 组 实 现 无 痕 删 除 我 们 应 用 该 方 法 连 续 删 除 了 大 肠 杆 菌 基 因 组 上 11 个 非 必 需 区, 使 基 因 组 减 小 10.59% 测 序 证 明 所 有 删 减 区 域 同 源 臂 未 发 生 突 变, 基 因 组 重 测 序 证 明 指 定 区 域 被 删 除 删 减 菌 的 生 长 变 化 不 大, 但 耐 酸 能 力 有 所 改 变, 并 对 番 茄 红 素 合 成 有 不 同 影 响 : 大 肠 杆 菌, 同 源 重 组, 无 痕 删 除 Received: February 10, 2015; Accepted: March 24, 2015 Corresponding author: Hongqing Fang. Some mutations in the fragments may be brought into the homologous regions near the target. To solve this problem in markeless gene deletion we developed a novel method characterized with two-step recombination and a donor plasmid. First, generated by a linear DNA cassette which comprises a I-Sec I site-containing marker gene and homologous arms was electroporated into cells for marker-substitution deletion of the target sequence. Second, after a donor plasmid carrying the I-Sec I site-containing fusion homologous arm was chemically transformed into the marker-containing cells, the fusion arms and the marker was simultaneously cleaved by I-Sec I endonuclease and the marker-free deletion was stimulated by double-strand break-mediated intermolecular recombination. Eleven nonessential regions in E. coli genome were sequentially deleted by our method, resulting in a 10.59% reduced genome size. These precise deletions were also verified by sequencing and genome resequencing. Though no change in the growth rate on the minimal medium, we found the genome-reduced strains have some alteration in the acid resistance and for the synthesis of lycopene.
