The core of the Smart Cycler System is the I-Core (Intelligent Cooling/Heating Optical Reaction) module, incorporating state-ofthe-art microfluidic and microelectronic design. Each Smart Cycler processing block contains sixteen independently programmable I-CORE modules, each of which performs four-color, real time fluorometric detection. May 2002 Cell-specific Genomics (cont. from page 1) Cell-specific molecular analysis of small cell populations captured by LCM typically requires isolation of RNA and amplification of the isolated amounts of RNA from nanograms to micrograms. Thus, the combination of LCM, RNA isolation & amplification, and microarray analysis or quantitative PCR reveals differential gene expression that might otherwise be obscured in whole tissue samples. Arcturus provides a complete range of dedicated products for preparing and staining of sections, for isolating DNA and RNA from captured cells and for linear amplification of the isolated RNA. RNA from only a few Cells The RiboAmp RNA Amplification Kit enables the production of large amounts of RNA from minimal starting quantities for use in gene expression studies. Amplification using the RiboAmp Kit expands the number of experiments that can be conducted from precious samples. Based on a proprietary, patent pending, process, mrna is linear amplified up to 1000 fold in one synthesis round and up to one million fold in two rounds. The RiboAmp system produces antisense RNA (arna), singlestranded RNA of complementary nucleotide sequence to the original mrna species. Successful preparation of micrograms of arna from low-nanogram quantities of total RNA enables routine studies of small cell populations prepared by Laser Capture Microdissection. In addition, amplification of mrna in all abundance classes ensures that differential gene expression patterns will be identified at all levels of original copy numbers. The PicoPure DNA and RNA Isolation Kits are optimized reagent kits for isolation of DNA/RNA from Laser Capture microdissected cells. Superior recoveries of nucleic acids from as few as 10 cells are consistently seen. Total RNA isolated using the PicoPure RNA Kit is provided in as little as 10 µl of buffer ready for use in the RiboAmp RNA Amplification Kit to produce microgram quantities of RNA suitable for labeling and hybridization in downstream experiments. Flexibility Speed Real time detection Four-color capability Expandability The Smart Cycler System Up to sixteen different cycling protocols can be performed simultaneously in one processing block. Experiments can be started at different times, allowing several operators to use the instrument concurrently. Rapid, precise heating and cooling significantly reduces cycling protocol time. Each of the sixteen sites are monitored in real time, growth curves are displayed and confirmation occurs at defined thresholds. Each of the sixteen reaction sites has its own four channel fluorescent detection. Up to four targets can be detected simultaneously. Up to six processing blocks of sixteen reaction/detection chambers can be controlled and monitored by one system. - 2 - Rapid DNA/RNA Amplification (cont. from page 1) The Smart Cycler software enables single or multiple operators to define and simultaneously carry out any number of separate experiments, each with a unique set of cycling protocols, threshold criteria, and data analysis. In addition, thermal and optical data from each and all sites can be monitored in real time, and graphs of temperature, growth curves, and melt curves can be charted as the data are collected. May 2002 PIPELINE A better way to dispense sterile fluids Essen Instrument's Pipeline was designed to replace tedious manual pipetting operations associated with high-volume cell culture protocols, such as in high throughput research applications. Pipeline provides a fast and accurate means for preparing multiple aliquots of a common reagent. Pipeline is also compatible with dispensing cells and suitable for use in cell culture. In multiplestep procedures, one can use an aspirator along with the Pipeline to exchange cell culture media, solutions and reagents without the need to change multiple pipettes. Two dispensing modes can be selected. The first mode dispenses a user-defined volume of fluid per switch activation, and the second mode dispenses continuously during switch activation. In addition, three different dispense speeds are possible allowing for the device to be tailored to the most demanding cell culture applications. The user has the ability to save and recall up to 8 pre-programmed settings, and the unit can be activated via front panel switch or foot switch. The accuracy and precision of the dispensed volumes are similar to serological pipettes. The Pipeline also provides an RS-232 interface if automation or remote operation is required. In addition to the -no wash- calcium release, drug resistance and membrane potential reagents for cell based screening, five kinase assay kits are now available. Proprietary IMAP technology developed at Molecular Devices is designed to evaluate enzyme activity of various protein kinases. Based on non-antibody fluorescence polarization (FP) detection method, phosphorylation of fluorescein-labeled peptide is monitored. The assays are a simple, homogeneous mix and read procedure that allows a fast and accurate determination of enzyme activity. The reagent kits incorporate kinase enzymes from Upstate Biotechnology, the leader in signal transduction. The principle of the IMAP technology is based on the high affinity binding of phosphate by immobilized trivalent metal ions on nanoparticles provided with the assay kits. This IMAP binding reagent complexes with phosphate groups on phosphopeptides generated in a kinase reaction resulting in change of molecular motion and increase of fluorescence polarization. Pyrosequencing Extends Genotyping System Capabilities New PSQ HS 96A and PSQ 96MA The new PSQ HS 96A (High Sensitivity) is a dedicated system for SNP and mutation analysis. It uses proven Pyrosequencing technology to enable real time detection of DNA sequencing events. Reliable chemistry and a simple, robust detection mechanism eliminate the need for gels, dyes or specific labels. A highly sensitive camera enables small amounts of sample DNA to be analyzed, typically 5-10µl of PCR product. Only small volumes of reagents and nucleotides are required, reducing the cost per accurate result. The system employs a 96-well plate format and is automatically fed, by a robot arm, with up to ten plates from the integrated stacker. The instrument analyzes up to 960 samples unattended per 2 hour run with a typical throughput of 4,500 samples in a working day, corresponding to a throughput of 30,000 polymorphisms in 24 hours using triplex multiplexing. New functionality include: simultaneous comparison of several pyrograms, automatic analysis of tri-and tetra allelic SNPs, automatic analysis of insertions/deletions (InDels), automatic analysis of multiple SNPs and multiplexing of polymorphisms such as point mutations, InDels and SNPs. Each polymorphism is analyzed using a separate sequencing primer. Also new: PSQ 96MA The PSQ 96MA System is a multi-application sequence analysis solution based on the original PSQ 96 hardware with new software and reagent kits that expand the range of applications. Internal control, provided by the sequence data, eliminates ambiguous results and the New PSQ HS 96A (High Sensitivity) need to reverify. Genetic variations can be analyzed with accuracy and confidence in a gel free operating environment. The results provide base sequence information, polymorphic position context and statistical analysis options.
