They are also the first to be registered and approved by the SFDA (State Food and Drug Administration). These thermal cyclers are programmable, feature rapid temperature change, accurate temperature control and can automatically record and analyze the data after amplification. Bioer has continued to progress and innovate its range and now introduces the LineGene 9600. This latest product in the Bioer real-time PCR detection system family is its first 96 well based system, with up to 8 detection channels and a greater temperature range than previous models. The unique patented block dissipation technology provide the LineGene 9600 with better heating and cooling rates and improved temperature accuracy, uniformity and stability. The LineGene 9600 uses dual colour scanning covering the whole fluorescence detection wavelength, to collect data from a 96 well plate in just over 5 seconds.
LineGene 9600 Real-Time Thermal Cyclers The LineGene 9600 series of instruments are 96 well Real-Time PCR systems capable of performing Absolute and Relative Quantification together with SNP and melt curve analysis. The LineGene 9600 is compatible with SBS footprint PCR plates, 8 or 12 strip-tubes and single 0.2ml tubes. This provides greater flexibility whether you are performing high throughput or low throughput applications. Being more versatile than high throughput specific systems, the LineGene 9600 series are ideally suited to meet the needs of any laboratory. The LineGene 9600 systems are equipped with gradient functionality for rapid optimisation, and have a melting temperature calculator useful for establishing protocols. There is a choice of block or tube temperature regulation. The latter uses an internal algorithm which calculates the temperature of the actual sample in real time during the PCR process. The hot-lid temperature can be set as required and the height adjusts automatically to the size of the tube or plate, effectively preventing evaporation. Technical Specifications Temperature This instrument is fitted with a peltier unit for thermal cycling with a temperature range of 4-105C. The incorporated patented block dissipation technology produces a ramping rate of 4C/second, allowing a run of 40 cycles to be completed in just over 60 minutes. The temperature uniformity of this block is on average ±0.3C and has temperature control accuracy of ±0.1C and a temperature fluctuation of ±0.1C. The block also has gradient functionality with a temperature span of 36C. Optics Excitation The LineGene 9600 series use long-lasting LEDs as the light source for the excitation of the individual fluorescence channels. LEDs work with very low energy and are stable in intensity and wavelength. Compared to other types of illumination they have a very long lifespan. System maintenance and calibration is minimised by the use of optical filter sets. Detection Photo-multiplier tubes (PMT s) are capable of detecting a single photon of light and demonstrate higher performance than CCD or photodiode systems. The LineGene 9600 series uses a unique dual photomultiplier detection system which measures the fluorescence signal from the bottom of the well to the top. This maximises signal detection and minimises well to well cross talk. Chemistry These open platform systems can be used with many different qpcr chemistries including hydrolysis probes and SYBR green based chemistries. Operation The LineGene 9600 systems can be run using a RS232C device, a USB device or an optional Bluetooth function connecting the instrument to a PC. Set up with Ease and Speed Software The LineGene 9600 series Research Software is included with the system. This easy-to-use intuitive software enables experiments to be set up with ease and analysed with minimal effort. It can be used for Absolute Quantification using a standard curve, Relative Quantification of different genes, together with Endpoint Genotyping analysis by detection of SNPs. Analyse with Ease and Speed User-friendly graphic programming, simple operation and practical. Temperature gradients can easily be determind using the Gradient Temperature calculator. The system has a gain calibration function enabling Auto gain or Manual settings. Analysis Data can be viewed and analysed quickly using the amplification plots, scatter plots, standard curves, auto call features and reporting options. Fluorescent and temperature curves are displayed in real-time, and conventional methods such as setting a threshold, linear regression or via second derivation are used to determine the Cq values. (Cq = quantification cycle, sometimes referred to as Ct = threshold cycle). Fluorescent curves can be shown in linear or logarithmic form as desired. The integrated quality system indicates if your results and controls are consistent with pre-defined parameters. The data can be exported to a.csv file, alternatively a customised report can be created.
Amplification Curve Example of a typical SYBR Green I amplification curve. The fluorescence signals of 96 samples are detected in each PCR cycle and shown in the graph. Baseline threshold, the maximum second derivative method and linreg3 methods are used to analyse the amplification curve of exponential growth of the start point, end point and closest point of efficiency, a key to automatically complete the analysis. The Analysis tab guides you through the data plots. The data can be further manipulated in Microsoft Excel. The reporting options can be customised to include appropriate data sets and other critical information. The reports can also be printed or easily exported into Microsoft Powerpoint. SNP Analysis Relative Analysis Melting Curve Post-amplification melting curve analysis is a simple way to check for specificity. Melting curve The graph shows the change in fluorescence plotted against the change in temperature, to provide a clear view of the melting dynamics.
