4 B I O O F O O D & F E E D S A F E T Y O V E R V I E W Food and Feed Safety Testing M A X I M I Z E S C I E N C E F O R L I F E 2 As one of the leading companies in the field, we develop, manufacture and market a wide range of food and feed safety test products covering Antibiotics/Veterinary Drugs, Hormones, Microbiological tests, Mycotoxins, Phycotoxins, and Scombrotoxin (Histamine). A majority of these determinations can be done by either HPLC, LC-MS or GC-MS; however, these traditional methods usually require very expensive preparation procedures, longer processing time, experienced technicians or radioactive materials. BIOO ELISA kits, on the other hand, provide a simple, rapid, sensitive and cost-effective screening method. BIOO MaxSignal TM ELISA test kits are developed and produced with BIOO proprietary antibodies and reagents to achieve high sensitivity and low detection limits. Our rapid, high recovery and cost-effective extraction methods are heavily tested using real samples from different regions of the world and constantly validated by world-wide reference labs. A BIOO ELISA kit contains one 96-well plate (8 wells x 12 strips) and reagents, including standards and buffers, required to perform the test. It has the capacity for 96 determinations or testing of 42 samples in duplicate. The shelf life is 12 months when the kit is properly stored. Procedures for sample extraction are described in detail in the manual. Antibiotics / Veterinary Drugs MaxSignal Antibiotics / Veterinary Drugs Test Kits are designed for the quantitative analysis of veterinary drug residues such as chloramphenicol (CAP), crystal violet/lcv, diazepam, enrofloxacin, fluoroquinolone, furaltadone (AMOZ), furazolidone (AOZ), gentamicin, malachite green/lmg, streptomycin, sulfamethazine, tylosin and other drug residues in animal feed, crab, eggs, fish, honey, meat, milk, milk powder, serum, shrimp, urine and other matrices. Hormones MaxSignal Hormone ELISA Test Kits are designed for the quantitative analysis of beta-agonists such as bitolterol, carbuterol, cimaterol, clenbuterol, diethylstibestrol (DES), ractopamine, salbutamol, zilpaterol and other hormones in animal tissue, animal feed, urine and other matrices.

5 Microbiological Tests Phycotoxins Phycotoxins are marine biotoxins that accumulate in fish and shellfish from their diet algae. There are three different types of phycotoxins: PSP (paralytic shellfish poisoning), DSP (diarrhetic shellfish poison) and ASP (amnestic shellfish poison). The consumption of phycotoxin-contaminated food can lead to serious health problems that can be recognized by the following symptoms: damaged nervous system, damaged digestive system and memory loss. Toxincontaminated mussels can be found worldwide in the summer time. Phycotoxins-residues can be determined by either HPLC, LC-MS or GC-MS. However, BIOO enzyme immunoassays (ELISA) have considerable advantages over conventional methods. Bacteria exist in a range of environments but impose danger to health when found in food and feedstuffs. Food products and their raw materials such as poultry meat, fish, shrimp, pork, and eggs can provide the perfect growth medium for microbes. For example, when Salmonella contaminated food is ingested it may result in the disease known as Salmonellosis. Clinical symptoms include vomiting, diarrhea and nausea; this is referred to as gastroenteritis and in severe cases may result in death. The detection of bacteria in food samples by conventional microbiological techniques is often difficult, laborious and time-consuming. Several days are required to culture the bacteria from a sample followed by a final isolation and identification by serological and biochemical techniques. BIOO enzyme immunoassays (ELISA), however, offer a faster method of identification as compared with traditional techniques. Mycotoxins Mycotoxin is a toxin produced by fungi, which includes mushrooms, molds and yeasts. It is a secondary fungal metabolite that induces a variety of toxic and carcinogenic effects when food contaminated with the toxin is ingested. There are two major kinds of fungi producing mycotoxins. One is Fusarium species, which generate mycotoxins directly on agricultural crops, such as DON, Zearalenone, T-2 Toxin and Fumonisin, predominantly reach the food chain through cereals and cereal products. The other important group of fungi (e.g. Aspergillus and Penicillium species) occur during inadequate storage areas. The corresponding mycotoxins, such as Aflatoxins and Ochratoxin A, are found in incorrectly stored crop products or in food products directly. USFDA and EU have set up a range of mycotoxins permitted in food and animal feed. Scombrotoxin (Histamine) Certain bacteria produce the enzyme histidine decarboxylase during growth. This enzyme breaks down free histidine, a naturally occurring chemical that is present in large quantities in some fish (Scombridae, such as tuna, kahawai, mackerel, bonito and butterfly kingfish). This results in the formation of histamine and other amines. Histamine fish poisoning (HFP) is frequently encountered if dead fish remain in set nets during warm sea temperatures, the fish is improperly refrigerated or when refrigeration is delayed. The symptoms of HFP mimic those of an IgE-mediated food allergy: nausea, vomiting, diarrhea, hives, itching, red rash (flushing), and hypotension. Malachite Green (Basic Green 4) has been widely used all over the world in the fish farming industry as a fungicide, ectoparasiticide and disinfectant. However, it has been shown that Malachite Green is highly cytotoxic to mammalian cells and also acts as a liver tumor-enhancing agent. This has led to a prohibition of Malachite Green for the treatment of fish and shrimp used for food production. To prevent Malachite Green residues from entering the food chain, both producers and government surveillance agencies need technologies and methods that provide rapid, accurate, and reliable results at the required sensitivity. Currently, Malachite Green residues are determined by either HPLC or LC-MS method with extremely expensive and time-consuming preparatory procedures. The advanced formulation enables users to deliver RNAi agents, in vivo, for target validation and drug target identification purposes. After formulating RNAi agents with MaxSuppressor TM In Vivo RNA-LANCEr, the RNA becomes stable in serum, preventing the need for stability modifications which reduces the synthesis cost. The preparation time takes less than one (1) hour and can be performed in any standard laboratory setting. Simply mix the RNAi agent with the semi-dry lipid cake of MaxSuppressor TM In Vivo RNA-LANCEr, sonicate and inject as shown in the figure below. However, the delivery route can be focused so that more efficient uptake and reduction may be obtained in your desired destination. Protamine sulfate is a small, positively charged, arginine rich protein which is traditionally used to reverse the anticoagulant effects of heparin by binding to it. It has been found that protamine sulfate can also associate with nucleic acids such as sirna and mirna molecules. For this reason protamine sulfate conjugated antibodies have significant potential to be used with RNAi agents for the directed delivery of these agents into animal tissues via cell surface receptors. To generate RNAi agent bound to protamine sulfate conjugated antibody, simply mix the RNAi agent with the conjugated antibody in the antibody storage buffer and incubate at 30 C for 15 minutes. Following the incubation the RNA/antibody complex can be diluted into a sterile saline solution and injected directly into the animal. This method is more economical than performing PCR in standard conditions and allows for a more physiological impact of mirna function a s compared with using PCR. ß-galactosidase ELISA Catalog # ß-galactosidase is a hydrolase enzyme that catalyzes the hydrolysis of ß-galactosides into monosaccharides. In E. coli the ß-galactosidase protein is encoded by the lacz gene which is part of the lac operon. The ß-galactosidase protein is a large (120 kda, >1000 amino acids) protein that forms a tetramer. The enzyme s function in E. coli is to cleave the disaccharide lactose to form glucose and galactose so they can be used as carbon/energy sources. The lacz gene from E. coli is one of the most commonly used reporter genes for testing the efficiency of expression vector mediated gene transfer and for studying the regulation of gene promoters. ß-galactosidase is not typically expressed in mammalian cells, however; ß-galactosidase positive staining is a marker for mammalian cellular senescence. ß-galactosidase is a useful reporter gene both in cell culture and in vivo applications due to its ease of detection. This ELISA kit contains all the reagents required to quantitate the level of ß-galactosidase protein in cells and tissues. This kit can also be used to study the influence of drug treatments on ß-galactosidase staining in animal organs or cells in culture. Luciferase ELISA Catalog # Luciferase is a generic name for enzymes, commonly found in nature, that produce light. The most commonly used Luciferase is produced by, Photinus pyralis, the firefly. Luciferase catalyzes a chemiluminescent oxidation reduction reaction and although many kits are available to detect the enzymatic activity of this protein using light emission, these methods have several drawbacks. These assays have rapid decay kinetics; require the enzymatic activity of the Luciferase to be retained and they do not enable absolute quantification.
