Microscope Selection How to Select the Right Microscope for Your Classroom Choosing the right microscope for your specific teaching needs can be confusing. Before you decide to buy, ask the following questions: Will you be teaching elementary, intermediate, high school or university students? Do you need a microscope with special features such as the ability to capture images or for other digital applications? Do you want a microscope that can be used in the classroom and out in the field? Do you want a microscope that can integrate with your existing technology or with future technology? Then evaluate the major microscope types: Compound Microscopes Comprised of two lens systems: the eyepiece and the objective. Provide high magnification power and a two-dimensional view of specimens. Have several eyepiece options: monocular (one), binocular (two) and trinocular (three, generally used with a camera) Total magnification power is equal to the powers of the eyepiece and the objective multiplied. For example, if a microscope has a 10X eyepiece and a 10X objective, the magnifying power is 100 times. Compound microscopes are ideal for applications such as viewing blood samples, cells and cell structures, bacteria, pond water and other liquids. Quality Microscopes for a Lifetime Ten Top Tips Towards Better Microscopy 1. A compound microscope gives a two-dimensional, flat image. Use a compound microscope for specimens to be examined using a slide preparation method (micro). There are monocular, dual-view, binocular and trinocular compound microscopes. 2. A stereo microscope has a binocular body and gives a three dimensional image. Use a stereo microscope for specimens that have depth or are large in size and require a working distance (macro). 3. Start to focus using the lowest magnification. In a compound microscope, this is the 4X objective. Make sure the objective clicks into place. In a stereo microscope, start by using the 1X. 4. Always place the specimen to be viewed in the center of the stage or stage plate. When using a compound microscope make certain the slide is placed on the stage with the center over the light. 5. When viewing a slide, make sure that it is right-side-up. This is especially important when using a prepared slide. If upside down, it will not be in focus on high power (40X)! 6. Focus first by using the coarse adjustment knob, then, the fine focus knob. You should be able to change from one objective (magnification) to another with just a minor fine focus adjustment. This means the microscope is parfocalled. 7. Adjust the illumination by using the intensity control and condenser or diaphragm. 8. Remember, when using higher magnifications, it is necessary to adjust the light source. 9. Teach your students the proper care and handling of the microscope. Always carry a microscope by using two hands; one hand around the arm of the microscope and the other under the base of the scope. 10. Keep your microscope clean. To clean the lenses, first remove any dust and dirt by using a camel hair brush or canned air. Moisten the end of a Q-tip with lens cleaning solution. Keep the other end dry. Clean the optical surface with the moist end of the Q-tip using a circular motion. Dry the surface with the dry end of the Q-tip using a circular motion. A solution of Windex with vinegar works well. Use a dust cover to store when not in use. 24

Quality Microscopes for a Lifetime Glossary of Microscopy Terms: Types of Objectives: Achromatic Objectives: Lenses which bring parts of the color spectrum into the same plane of focus. Achromats are standard on any quality microscope. They may be designated as flatfield objectives but typically deliver between 60-70% flat field. Semi-Plan Objectives: Objectives that are corrected to attain a flatness of field around 80-85%. Plan Achromats: Objective lenses delivering nearly 100% flat field. Phase: Objectives create the effect of staining by changing the optical light path without actually staining the slide. Benefit is that specimen remains viable and intact. Basic Definitions: Chromatic Aberration: When lenses fail to focus two different parts of the spectrum in the same focal plane, they suffer from chromatic aberration. Coaxial Controls: The focusing control mechanism moves the stage up and down to bring the specimen into focus. A coaxial focus control combines the coarse and fine focus mechanisms into one control. This coaxial focus control incorporates a clutch mechanism which allows for slippage at the extreme ends of the focus range to prevent damage to the gears. Condenser: The function of the condenser is to provide full illumination to the specimen place and to enhance the resolution and contracts of the object being viewed Depth of Focus: The ability of a lens to furnish a distinct image above and below the focal plane. Depth of focus decreases with the increase of numerical aperture or with the increase of magnification. DIN Optics: A German Standard for the manufacturing of microscope lenses. Optics are interchangeable from one DIN microscope to another. (Deutsche Industrial Normen) Diopter Adjustment: The ability to adjust the focus for one eyepiece in a binocular or trinocular microscope to compensate for the difference in vision between the user s eyes. Focal Length: Parallel rays of light after refraction through a lens will be brought to a focus at the focal point. The distance from the optical center of the lens to the focal point, in the focal length, or focus. Iris Diaphragm: A device mounted underneath the stage which slides open to accommodate light up onto the stage opening. Numerical Aperture (N.A.): A measure of the light gathering capabilities of an objective lens. The concept is comparable to the F-value in photographic lens. In general, N.A. values less than 1.00 are dry objectives, while values greater than 1.0 require oil as a medium. The N.A. value can be found on individual objectives. Note: Condenser lenses are part of the optical system and are also assigned a N.A. Value. The condenser system on a scope should match the N.A. of the highest power objective on the microscope. Oil Immersion: When light rays from the objective lens to the specimen pass through air, they are distorted slightly. This is not a perceptual problem at a magnification of 400 or lower. However, at a magnification of 1,000, the slight distortion is reduced significantly by placing a thin layer of very clear but viscous oil between the slide and the tip of the objective lens. Parfocal/Parcenter: A microscope system that is parfocused/parcentered enables the user to switch objective lenses (change powers) and still have the specimen in focus and centered in the field of view. Resolving Power: The ability of a lens to reveal the fine details in a specimen. Poor quality lenses suffer from empty magnification because they enlarge the subject with out resolving additional detail. Retractable Lens: A specially designed lens which the tip retracts on contact to prevent damage to the slide or to the objective. Typically found on higher magnification objectives: 40X, 100X. Working Distance: The distance between the front lens of the objective and the cover glass when the lens is focused on the specimen. 26
Glossary Illumination Options: Tungsten Incandescent: Standard tungsten filament bulb. Widely available and inexpensive. Produces a bright light, but is very hot. Not recommended for viewing live specimens. Halogen: Halogen bulbs are very bright, and produces heat. Not recommended for viewing live specimens. LED: Light emitting diode which produce a very cool and white light. Energy efficient and long lasting. Digital Terms: Exposure: Refers to how long the shutter of the camera is open, not to brightness. Field of View: The image that is seen through the microscope eyepiece may differ from the image that is seen on the computer screen. The quality of the microscope affects the quality of the image. Frame Rate: The measurement of the frequency or rate at which a digital camera produces images called frames. Frame rate is usually expressed in frames per second. (FPS) This is important in video streaming. Megapixel (MP): Refers to the number of pixels in an image and to express the number of sensor elements of a digital camera or the number of display elements of a digital display. For example, a camera with an array of 2048X1536 pixels is commonly referred to as 5 MP camera. Resolution: Live image resolution is most important in digital microscopy. This may be different than what is used in digital photography. When using a microscope as a teaching tool, it is important to see what is happening under the microscope. Live image resolution is the part that allows one to see a smooth curved cell wall without pixilation. It is important to choose the right image chip that will give the right live image resolution.
