MACHEREY-NAGEL Company profile Trusted quality since 1911 Since 1911 MACHEREY-NAGEL has established itself as a company for high quality, innovation, and reliability in biomolecular and chemical analysis. Friendly expert advice for our highly valued customers as well as outstanding product quality (EN ISO 9001:2008 and EN ISO 13485:2003 certified) have been the cornerstones of our corporate success for over 100 years. MACHEREY-NAGEL is a family-owned business run by the fourth generation, the great granddaughter of the company s founder, has been managing the enterprise since 2000. The companies headquarter is located in Düren, Germany. Today, the complete product portfolio includes Bioanalysis, Filtration, Rapid Tests, Water Analysis, and Chromatography. The Bioanalysis department provides innovative, client-driven solutions for DNA / RNA purification, clean-up, and concentration. Forensic quality products Consumables used in forensics need to be treated carefully to prevent DNA contamination. MACHEREY-NAGEL therefore has a stringently controlled production process to avoid DNA contamination of consumables. Further, MACHEREY-NAGEL uses ethylene oxide (EO) treatment to remove amplifiable DNA, which might still be introduced during the manufacturing process. MACHEREY-NAGEL products carrying the forensic quality seal, contain plastic materials that are EO treated. This means, DNA of any kind, which might still be introduced into plastic consumables during the production process, is inactivated by means of the treatment with EO, in order to prevent the generation of accidental human profile by PCR amplification. EO treatment has been shown to be the method of choice to prevent DNA profiles due to DNA contamination. NucleoSpin Forensic Filters Sample preparation Spin basket solution for lysis incubation and subsequent separation of lysate from samples such as swabs, denim, cigarette butts, etc. XXLysis and separation in one tube no sample transfer, no extra pipetting steps XXSpecial filter design absolutely leak proof, compatible with all typical lysis buffers XXComplete DNA recovery and process reliability, no DNA loss, no cross-contamination Technology Sample matrial Maximum volume capacity Forensic quality Special semi-permeable baskets Swabs, denim, cigarette butts, and other sample carriers 800 μl Ethylene oxide treated NucleoSpin Forensic Filters are the simplest and cleanest solution for lysis incubation and post-lysis separation of lysates from substrates. This unique product is especially designed for the forensic market. The NucleoSpin Forensic Filters enable the incubation of any solid sample carrier with any lysis buffer for any amount of time without leakage. Only a centrifugation step > 1 min at 10.000 x g separates efficiently the solid carrier from the lysis buffer containing the desired DNA. The NucleoSpin Forensic Filters ensure a complete DNA recovery even from sample with only traces of DNA, without any cross-contamination. Prepare Incubate for lysis Spin for separation New product Processing swabs with the NucleoSpin Forensic Filters results in a significant increase of DNA yield! Product evaluation revealed up to 60 % higher recoveries in comparison to standard procedures. References and customer testimonials will follow shortly. NucleoMag Forensic DNA extraction DNA isolation for forensic sciences reliable quality for your STR analysis XXExcellent DNA purity from all types of casework samples XXMagnetic bead based isolation XXHigh sensitivity for low and high DNA content Technology Sample material Typical yield Elution volume Binding capacity Magnetic-bead technology Casework samples, contact traces (e.g., dried blood spots, cigarette filters, swabs) Depending on sample amount and quality 20 25 μl 0.4 μg/μl beads The NucleoMag Forensic kit is designed for the reliable purification of DNA from different casework samples in forensic science (e.g., buccal swabs, blood spots, cigarette butts, chewing gum, epithelial cells, body fluids on fabric). The kit provides high quality DNA for your STR analysis from samples with high and low DNA content. The kit uses a magnetic bead based purification procedure, which can be used manually or automated on liquid handling platforms. The special lysis buffer allows the direct processing of different sample materials. Samples on a carrier should be separated after lysis using NucleoSpin Forensic Filters. DNA yield strongly depends on the DNA content in the specific sample (typical yield from buccal swab is 400 800 ng). The extracted DNA can be used directly as template for PCR. The kits can be used manually under centrifugation or vacuum as well as on automated common liquid handling instruments. NucleoSpin 8 / 96 Trace Strip / Plate Customer testimonial The Landeskriminalamt Berlin (Berlin Crime Investigation Authority) is using NucleoSpin 8 / 96 Trace (MACHEREY-NAGEL) for extracting genomic DNA from forensic samples since 2005. We achieve satisfactory results with complete STR profiles from approximately 94 % of all buccal swabs in the first run and from another 4 % of all swabs by using a larger volume of the elution fraction in a second PCR. Up to now (August 2008) the throughput of buccal swab samples was in a range of more than 30,000 samples. Besides buccal swabs, DNA is also extracted from cigarette filters (more than 10,000 samples) and other trace materials by using the same system.
