3 Advanced Filtration Ahlstrom GenCollect Sample Collection s Ahlstrom s GenCollect card is an easy and cost-effective way to collect and store biosamples at ambient temperature for a wide range of applications such as genotyping, forensics and paternity tests. Key features Ahlstrom cards, made of high purity cellulose, provide a rapid and safe way to collect, transport and store biological samples at ambient temperature. Nucleic acids can be easily extracted from the biosamples for subsequent analyses. Ahlstrom offers GenCollect, a white card format and Ahlstrom GenCollect Color, a pink card format. Ahlstrom GenCollect card is recommended for easily identified samples, such as whole blood, while Ahlstrom GenCollect Color is useful for clear samples such as saliva, buccal cells or urine. 4 Simple Collection Easy Extraction Reliable Analysis Upon application of biosamples to the Ahlstrom GenCollect and Ahlstrom GenCollect Color cards, nucleic acids become preserved for short-term storage at room temperature Quantification and STR analysis of DNA purified from Ahlstrom GenCollect and competitor s cards DNA quantification Real-Time PCR amplification was performed to assess the quantity and stability of genomic DNA purified either from human blood collected on GenCollect (Ahlstrom) and competitor s card or from saliva collected on GenCollect Color (Ahlstrom) and competitor s pink card. After collection of samples, cards were dried at room temperature for a minimum of 3h. Immediately after the drying step or after 60 days of storage at ambient temperature, DNA was extracted from discs punched from the cards and purified using Crime Prep Adem-Kit: Casework according to the manufacturer s protocol (Ademtech). Recovery of genomic DNA stored dry at room temperature on Ahlstrom GenCollect cards indicates an improved sample protection and a higher yield of recovery as compared to competitor s cards. 5 Advanced Filtration STR genotyping analysis STR analysis was used to evaluate the quality and purity of genomic DNA purified from saliva spotted on Ahlstrom GenCollect Color card. The biosamples not stored or stored at ambient temperature for 60 days were subjected to amplification. Both sample types showed strong signals and high quality peaks. A full profile with all STR loci (24) amplified was obtained and no inhibition was observed. Same result obtained with Ahlstrom GenCollect card. Extraction and purification of DNA from Ahlstrom GenCollect Color card provides DNA with sufficient quantity and high quality to support allele calling accuracy as high as 100% in STR analysis. STR profiles from DNA purified from discs punched from 7 μl of saliva collected on Ahlstrom GenCollect TM Color a) not stored or b) stored for 60 days at ambient temperature. Both samples were processed using a GlobaFiler TM PCR Amplification Kit. The samples were run on a 3500xL Genetic Analyzer (Applied Biosystems) using standard conditions and analyzed using GeneMapper ID-X V1.4 (Applied Biosystems).
