1 Genera Certificate of Education Advanced Subsidiary Examination June 2013 Human Bioogy HBI3T/Q13/task Unit 3T AS Investigative Skis Assignment Task Sheet The effect of sodium choride concentration on the time taken for the reaction cataysed by amyase Introduction Starch is a poysaccharide that reacts with iodine soution to produce a bue-back coour. Amyase is an enzyme that catayses the digestion of starch. When amyase is added to a soution of starch, the starch is digested over time. A the starch has been digested when a sampe of the soution no onger gives the bue-back coour when mixed with iodine soution. Sodium choride affects the abiity of amyase to digest starch. You wi investigate the effect of different concentrations of sodium choride on the time taken for the digestion of starch by amyase. Materias You are provided with: amyase soution starch in ph7 buffer soution sodium choride soution, concentration 2.0 mo dm -3 distied water iodine soution water baths or arge beakers that you can use as water baths thermometer spotting ties test tubes test-tube rack or arge beaker to stand test tubes in timer marker pen or Chinagraph penci graduated pipettes or syringes dropping pipettes. You may ask your teacher for any other apparatus you require.

2 2 Outine method Read these instructions carefuy before you start your investigation. Your teacher wi suppy resuts for tubes D and E. 1. Labe three test tubes A to C. 2. Add the reagents shown in the tabe to each abeed tube. Use a cean graduated pipette for each soution, starting with the starch. Soution Voume of soution in each tube / cm 3 A B C D E Starch in buffer at ph mo dm 3 sodium choride soution Distied water Stand a three tubes (A to C) and the container of amyase in a water bath at 50 o Cfor at east 10 minutes. 4. Whie you are waiting, prepare the spotting ties by putting one drop of iodine soution in each we. 5. Use a cean pipette to add 3 cm 3 of amyase soution to tube A. Mix the contents of tube A and return it to the water bath. 6. Start the timer. 7. Immediatey, use a dropping pipette to add 1 drop of mixture from tube A to a we of iodine soution in the spotting tie. Return any unused mixture to the test tube afterwards. 8. After every 30 seconds, add a further drop of mixture from tube A to a new we containing iodine soution. Return any unused mixture to the test tube afterwards. 9. Repeat Step 8 every 30 seconds unti the reaction of the iodine soution shows that a the starch has been digested. 10. Record the time taken for a the starch to be digested. 11. Cean the spotting ties and repeat Steps 4 to 10 using tube B, then C. You wi need to decide for yoursef: how many repeats to carry out when the coour of the soution in the we shows a the starch has been digested.
