Performance QC and Assay Seup Performance QC Open he Seup & QC workspace. Selec Seup & QC. Verify he bead lo ID. Click Sar. Run he BD FACSuie CS&T Research beads. View he Seup and QC Repor and QC Tracking abs. Verify ha he Performance QC passed. Assay and Tube Seings Seup Selec he Seup & QC ab. Selec he Assay & Tube Seings Seup ask. Click Selec. Acquire Daa Open he Experimen workspace. Selec Acquire Daa In Experimen. Selec File > Rename o rename he experimen. Creae plos, gaes, saisics, and a hierarchy needed for acquiring daa. Use he Workshees oolbar o creae plos, gaes, saisics, and a hierarchy. Righ-click a plo and selec Properies o modify i. Insall a sample ube ono he cyomeer. Click Preview. Click PMTV o enable he sliders in he workshee o make adjusmens o he PMT volages, as needed. Or, apply he appropriae ube seings. See Sep 5. Verify ha he volage and hreshold seings in he defaul ube seings are appropriae. Verify ha he gaes and populaions are se appropriaely. 4 Click Sop. -0-00 Rev. 0
5 Double-click he ube o modify he ube properies. Rename he ube. Click Selec o apply differen ube seings as needed. Add or remove fluorochromes as needed. Change fluorochromes as needed. Clear any A, H, or W measuremens no needed. Specify reagen labels, keywords, and acquisiion crieria as needed. 6 7 Click Nex in he Daa Sources panel o creae addiional ubes, as required. Analyze Daa Creae any addiional plos, gaes, and saisics in he workshee, or creae a repor. Cusomize plos by righ-clicking a plo and selecing Properies. Perform qualiy conrol of he analysis. Verify ha gaes are se appropriaely for all samples. Use he populaion hierarchy o verify paren/child relaionships. Do one of he following o prin or expor he resuls. Selec File > Prin o prin he acive workshee. Selec File > Expor o PDF o creae a PDF of he acive workshee. Shu Down he Sysem Selec Cyomeer > Daily Clean. Selec Cyomeer > Shudown. Addiional Informaion Any experimen can be used o creae a user-defined assay and used in a worklis o acquire and analyze daa. Addiionally, experimens can be creaed from assays and used o acquire and analyze daa in he Experimen workspace or saved as an assay o be used in a worklis. Creaing an Assay from an Experimen Open an exising experimen or creae a new one. Selec File > Creae Assay. Ener a name. Selec a repor o display, if needed. Click OK. Creaing an Experimen from an Assay Selec File > New Experimen from Assay. Modify he assay as needed. Selec File > Creae Assay o save he experimen as an assay and acquire daa in a worklis.
