5 Sample and Substrate Flexibility Protein array technology has expanded beyond the simple printing of purified antibodies to include more complex samples such as cell lysates and tissue homogenates. Our patented deposition technology, combined with our many years of experience with reverse phase lysate arrays (RPLA), enables us to produce cell and tissue lysate arrays of exceptional reliability and quality. Our SearchLight Protein Array Technology is a multiplexing sandwich-elisa system based on chemiluminescent or fluorescent detection of analytes whose respective captureantibodies are spotted in arrays within each well of a 96-well microplate. As many as 16 analytes (4 x 4 array in each well) can be measured per well, meaning that 16 cytokines or other biomarkers can be assayed simultaneously with each 50 µl sample! 9 The SearchLight Sample Testing Service Our biomarker sample testing service gives you the flexibility to select only your biomarkers of interest from our growing menu of more than 300 biomarkers. The biomarkers requested are grouped into arrays based on compatibility and anticipated expression levels. Each sample is tested in duplicate at appropriate dilutions in custom SearchLight biomarker array kits. A confidential Analysis Report (figure 1) displays the concentration of each analyte in the array for each sample tested. Values reported are the mean value for each sample at the optimal sample dilution. With help from experienced Aushon scientists, you choose from our menu of more than 300 biomarkers to design the unique panels that are most relevant to your specific research project or therapeutic area. Biomarkers are grouped based on anticipated expression levels and carefully screened for specificity. Individual array panels can be formally validated for pre-clinical and clinical studies, while new biomarker assays can be developed and incorporated into a custom array panel. 11 Custom Array Validation Large studies or trials may require validation of the specific array panels (combinations and configurations of analytes) that will be used for sample analysis. Assay protocols, sample types tested, and specifications for the validation assays can be modified to meet individual study requirements. Aushon researchers have validated specific biomarker array panels for a number of clients for Alzheimer s, cancer, cardiovascular disease, COPD, cystic fibrosis, diabetes / obesity, inflammation, rheumatoid arthritis (RA) and other studies. Each new antibody pair is tested with natural samples to confirm proper recognition of natural protein. The Aushon SearchLight Plus Imaging and Analysis System is designed specifically for quantitative chemiluminescent protein arrays. The CCD Imager offers a sleek desktop design and features rapid imaging (typically <2 minutes per plate) and a fixed-focus setting. Once the instrument is installed, no focus adjustments are required from one image to the next making it easily shared by multiple users. Plus, the automated calculation of optimal image time makes the instrument incredibly easy to use. Specialized array software is included for accurate, reproducible analysis of SearchLight Protein Array images.
