3 EASY AND INTUITIVE SOFTWARE K1-Fluo s dedicated software controls the confocal microscope with easy and intuitive user interface. The software provides many useful functions such as ROI (region of interest) scanning, navigation window, and various scanning method which includes simultaneous or sequential detection. It includes line profile, intensity time chart, and additional image processing functions. It also supports various save options and image file types. The advanced functions are included and developing; delicate ROI photobleaching or photoactivation, FRAP, and FRET function. 3D sectioning is realized with automatic Z stage or automated microscope body. We are happy to fully support third-party automation instruments. Software engineer is readily available for custom software; for first-stage research or prototyping. 4 Multi Channel Detection Highly sensitive standard detectors and multi-wavelength diode lasers enable imaging of multi-labeled specimen. Various combinations of laser and detector are available with optimized filter combinations. Optional low light detector module is also available for low light or high speed applications. The ultra-high sensitive detector offers high quantum efficiency and low noise with expense of cost. Line Profile Graph The digitized images can be directly used for quantitative analysis. By drawing the line on the Display Windows, the line profile graph is popped up to show numerical data such as intensity profile along the line. *BPAE cells with mouse anti- -tubulin Intensity Time Chart Intensity signal variation by lapse of time is displayed as time chart. Intensity of the selected area is calculated and plotted at every moment. *BPAE cells with mouse anti- -tubulin 5
5 Navigation System By drawing the ROI (region of interest) box in the Navigation Window, the laser scanning area can be accurately defined. Display Widows shows the magnified images by scanning the boxed area only. It actually provides the continuous optical zoom for selected ROI. *Muntjac cells Array Scan (Image Stitching) The consecutive imaging of pre-defined area, and stitching of these acquired images will provide a lot of convenience for the wide range imaging of large samples. The stitched image can be analyzed as one single image, and/or the user can also observe original individual images to see specific segmentations of the stitched image in detail. Slide glass imaging, or well-plate scanning is available with this function. 416 μm 2080 μm *BPAE cells with mouse anti- -tubulin (imaging by 20x objective lens) 6
6 Time Lapse Time-variant images can be acquired at various frame rate by Periodic scan function. Image acquisition at user-defined time interval enables the analysis of changes over time. Both simultaneous detection and sequential detection modes are available. 3-Dimensional Display 3D optically sectioned images can be acquired with automatic vertical motion of Z stage or automated microscope body. Z-step interval, pinhole size, and image pixel can be controlled for optimal condition. Display of 3D reconstructed image shows more information to researchers. Various display options are available. ROI Photobleaching the specific area is a very useful technique. This is useful for studying mobility of proteins with analysis method such as FRAP (Fluorescence Recovery After Photobleaching) or FLIP (Fluorescence Loss In Photobleaching). *BPAE cells with mouse anti- -tubulin 7
7 OPTIONS XY Stage High precision and accuracy manual and motorized stages are ready for every commercial microscope body; Nikon, Olympus, Zeiss, Leica, upright and inverted microscope. Various specimen holders including glass slides, multi-well plates, Petri-dishes are available. Z Focus Motor The manual type z-focusing stage of a microscope can be automated in two different ways. The step motor focusing stage is most suitable for the general microscope body, which encloses and rotates the z-focusing knob directly. Step sizes as small as 0.1μm gives precise focus and repeatable positioning in the Z-axis. High precision PZT stage gives extremely high accuracy and small step size of 0.1nm, but its stroke is limited to 400μm and it is only for single objective lens. Microscope Body Every manual or automated microscope body is adaptable with fully automated software. Used microscope is also adaptable after tiny modification. Laser Wavelength Up to four stable diode lasers are selectable among commercialized laser wavelengths. Please refer specification table. CCD Various CCD types are adaptable to most of microscope bodies. CMOS, cooled CCD, large format CCD, color or monochrome CCD is also available. Objective Lens Every maker s objective lens is available. We recommend highly corrected lens for confocal microscope. Low Light Detector (Ultra-high Sensitive Detector) Ultra-high sensitive detector is adaptable instead of standard detector for low light or high speed application. This detector offers specialized quantum efficiency and low noise character with expense of cost. 8
8 DMB Automated Digital Microscope Body *K1-Fluo with DMB DMB is a compact-size automated microscope body with 3-axis (x,y, and z) motorized stages, a turret, and imaging optics, by removing eyepieces and lamp illumination. K1-Fluo is compatible with every commercial microscope body, however, DMB can replace a commercial microscope body. Designed to be easily modified for automated imaging and data recording functions, it maximizes the convenience of operation and performance of K1-Fluo.
