6 SORBENTS Sample preparation and Clean-up are the key steps in trace analysis for analytical chemist. Solid phase extraction (SPE) is a powerful technique to provide a rapid and selective sample clean-up, a high recovery and the concentration necessary for accurate quantitative analysis. A large number of phase chemistries is available and can be processed using dedicated vacuum manifold accessories. A multitude of phase chemistries and formats (SPE tubes, 96-well plates,.) are available in this catalog. AFFINIMIP SPE for high affinity extraction of analyte from complex matrices and ready to use protocols. AFFINIMIP SPE AFFINIMIP SPE is a selective solid phase extraction based on Molecularly Imprinted Polymers (MIP) that combines the advantages of immune-affinity columns regarding the selectivity and those of polymeric SPE sorbents in terms of costs and robustness. Developed for very problematic analyte purification, an instruction sheet gives all required information to successfully carry out the analysis, including the protocol. No method development is required. The protocol is easy, simple and fast. The chromatogram analysis is fast thanks to the affinity between the analyte and AFFINIMIP SPE. Methods are developed for the usual analyte containing matrices. These matrices can be as diverse as aqueous matrices (urine, serum), fully organic matrices (oil), powder (milk) or cereals grains. If your sample preparation is complex or gives inappropriate results, the development of an AFFINIMIP SPE can be a solution. Moreover, POLYINTELL has developed unique products for radiopharmaceutical applications (Catalog for radiotracers purification). AttractSPE TM Polymeric sorbents AttractSPE TM polymeric sorbents are the last generation of SPE sorbent. They are crosslinked polymer particles bearing various chemical functionalities. A limited number of sorbent (6 products) simplifies your choice and covers the full range of analysis using aqueous matrices. These sorbents provide all the advantages of polymeric sorbents, including robustness (to solvent, ph and T), simplified method development, wide applicability and not affected by drying out. The combination of the water-wettable optimised surface chemistry, high surface area and ph stability ensures high reproducible recoveries for a wide range of analytes. For the same application, less sorbent and less solvent are required than for silica-based sorbent and so, their added value is particularly obvious for trace analysis and for small volume sample. Silica-based sorbents The first sorbents historically used for SPE applications, Silica-based sorbents offer a broad range of chemically modified sorbents. This chemistry goes from very polar sorbent (bare silica) to hydrophobic sorbent (endcapped saturated hydrocarbon modified silica) passing through intermediate polarity (for instance, amino modified silica). These sorbents are sensitive to extreme ph. 6

7 SOLID PHASE EXTRACTION SPE PROCEDURE STEPS Solid phase extraction requires the following steps: 1- Sample preparation: This step is necessary to obtain a loading solution compatible with retention condition in the SPE column. In case of solid matrices, this solution must also extract analytes from these matrices. 2- Conditioning: The SPE cartridges are conditioned with the appropriated solvents to fully wet sorbent and enable further interactions between the analytes and the sorbent. 3- Loading: the percolation solution is loaded through the SPE cartridge. The analyte must be retained in the column as well as unwanted compounds. 4- Washing: interferents and unwanted compounds are washed off by using several solvents. 5- Elution: The desired analyte is extracted from the SPE cartridge. Loading Washing Elution Analytes of interest Retention Mechanism SPE cartridges are available in a complete and wide range of sorbents. A sorbent is often classified according to the main retention mechanism between the analyte and the sorbent (normal phase, reversed phase, ionexchange, mixed-mode). Normal phase retention mechanisms are based on polar-polar or dipole-dipole interactions between the analyte and a silica-based sorbent. Reversed phase retention mechanisms are based on non polar- non polar interactions and Van der Waals dispersive forces. The sorbent is hydrophobic (polymeric sorbent or alkyl or phenyl modified silica-based sorbent). Ion-exchange retention mechanism use electrostatic interaction between a charged sorbent and the ionic analyte. The sorbent is charged with the opposite charge of the analyte. A cationic (respectively anionic) analyte interacts with a cation (respectively anion) exchange sorbent. Mixed-mode sorbents mainly interact through reversed phase and ion exchange retention mechanisms. They are available as a polymeric sorbent (AttractSPE TM SAX, WAX, WCX or SCX) or as SiliaPrep. 7

8 QUICK LOOK UP GUIDE The choice of the sorbent is crucial for a successful sample pre-treatment. When AFFINIMIP SPE exists for the analyte, the choice is very simple and the problem is solved. The protocol is ready to use. AFFINIMIP SPE have been developped for very problematic analysis with common SPE sorbents. When no AFFINIMIP SPE product exists, the sorbent chemistry must be chosen very carefully and each step of the protocol must be optimized. This choice is based on the nature of the analyte and of the matrix, on the loading condition and on the planned retention mechanism with the sorbent. The following scheme gives a brief guideline to select the family of sorbents. For specific applications such as trace analysis or complex matrices, conventional SPE sorbents may give very complex analysis or unsatisfaying results. The development of an AFFINIMIP SPE sorbent for this application is a solution. 11 AFFINIMIP SPE FOR ANALYTICAL PURPOSE AFFINIMIP SPE is a selective solid phase extraction based on Molecularly Imprinted Polymers (MIP) that combines the advantages of immune-affinity columns regarding the selectivity and those of a classic Solid Phase Extraction (SPE) in terms of costs and robustness. The advantages of AFFINIMIP SPE are essential in trace analysis from a complex matrice in food safety, environment and pharmaceutical analysis such as Mycotoxins (Patulin, Ochratoxin A, Zearalenone), phenolics, endocrines disruptors (estrogens), Bisphenol A, GSH adducts, etc. AFFINIMIP SPE Selective Extraction Cartridges Perfect clean-up system for trace analysis Thanks to selectivity of AFFINIMIP SPE, stringent washing steps can be applied in order to remove all interferences and thus minimize matrix effects. It also reduces ion-suppression effects. The SPE protocol is supplied in an instruction sheet for various complex matrices. Minimal to no method development required A protocol based on three steps (loading, washing and elution) is supplied with AFFINIMIP SPE kits for tested matrices. No extra-equipment than the usual required for SPE experiments is necessary. AFFINIMIP SPE protocols are as well defined by 3 steps of loading, washing and elution. 12 AFFINIMIP SPE VS IMMUNOAFFINITY COMPARATIVE STUDY Solid phase extraction is a simple tool to selectively extract analytes from complex matrices and quantify concentrations lower and lower. The major disadvantage of conventional SPE sorbents, such as C18 is a lack of selectivity and interference matrix components are co-extracted with the target analytes. To solve this problem, affinity-based SPE sorbents have been developed to be selective in extracting the target analytes like molecularly imprinted polymer (MIP) and immunoaffinity sorbent. Immunoaffinity columns (IAC) are biological sorbents based on the use of antibodies that are specific to the target analytes. Molecularly imprinted polymer is a synthetic material with artificially generated three-dimensional network able to specifically rebind a target molecule. Based on molecularly imprinted polymers, POLYINTELL s AFFINIMIP SPE cartridges have the advantages to be highly selective and specific. Contrary to IAC, AFFINIMIP SPE cartridges are chemically and thermally stable, compatible with all solvents as well as cost effective. PET commonly utilizes the radioactive forms of carbon ( 11 C), nitrogen ( 13 N), oxygen ( 15 O) and fluorine ( 18 F). The use of these isotopes allows the labelling of many different substrates without altering their biological activity. The half lives of some nuclei are relatively short, which poses a timescale problem for radiochemists and can leave little room for manoeuvre between introducing the radioisotope into the tracer, and conducting the PET scan. Of these isotopes 18 F has the most convenient half life of minutes.
