1 Application Notes # LCMS-35 esquire series Application of LC/APCI Ion Trap Tandem Mass Spectrometry for the Multiresidue Analysis of Pesticides in Water An LC-APCI-MS/MS method using an ion trap system was developed for the specific, sensitive and simultaneous quantification of seven pesticides (carbamates, triazines and phenylureas) in water samples. Detection limits down to 0.1 pg/µl (5 pg abs.) were achieved, together with high linear ranges and good reproducibilities for multiple analyses. Introduction The benefit of liquid chromatographic/ tandem mass spectrometric (LC-MS/ MS) methods for environmental analyses like quantitation of pesticide residues in various matrices is well known [1]. Compared to UV detection, these methods generally allow for enhanced specificity and lower detection limits. Furthermore, the ionization techniques used for LC-MS are known as soft ionization techniques, so that even very polar or thermolabile compounds (as for example carbamates) can be handled, which otherwise are difficult to analyze. For quantitation tasks, triple quadrupole systems are often used as mass spectrometric detectors, wheras ion trap systems are not that wide-spread, because they often are thought to be less sufficient in terms of detection limits, accuracy and reproducibilty. Nevertheless, ion trap systems show clear advantages if full scan sensitivity, sophisticated experiments or high scan speed with the possibility of fast switching between different experiments is required, as for example in the case of proper peak form recording for coeluting compounds. Therefore, in this work the suitability of an LC/ion trap MS system for the simultanous quantitation of a multiresidue pesticide mix was evaluated. Experimental Conditions HPLC Conditions Instrument: HP 1100 HPLC (Agilent) Column: XTerra C µm, 3.0 x 100mm (Waters) Mobile phase: gradient; A: water (0.1 % HCOOH), B: acetonitril Gradient: min: isocratic 5 % B (v/v) 1 10 min: gradient from 5 % B to 25 % B min: isocratic 36.2 % B min: gradient from 36.2 % B to 100 % B min: isocratic 100 % B min: gradient from 100 % B to 5 % B > 35 min: isocratic 5 % B Flow rate: 0,5 ml/min Temperature: 25 C Samples: Standard: mix of all seven pesticides, concentration levels 0.1, 0.5, 1, 5, 10, 50, 100, 500, 1000, 5000 pg/µl. Samples: blank surface water, blank drinking water, spiked surface water, spiked drinking water Inj. volume: 50 µl; 7 injections for calibration levels of 5 and 100 pg/µl, 1 injection for the other calibration levels, 3 injections for samples 8 and 9, 7 injections for samples 10 and 11 MS Conditions Instrument: esquire3000 plus Ion Trap MS system (Bruker Daltonics) Source: APCI positive, APCI temperature 200 C Scan: m/z MS/MS mode: 8 spectra averaged for each aquired data point; more ions mode: 4 spectra averaged for each aquired data point Mode: standard normal (resolution 0.6), MS/MS mode MS/MS and tune parameters set individually for each compound via time segment definitions: 0 9 min: MS/MS 163 ([M+H] + Methomyl) min: MS/MS ([M+H] + Desethylatrazin) min: MS/MS 157 ([M+H] + Aldicarb) min: MS/MS and MS/MS ([M+H] + Atrazine and Chlortoluron; mode more ions : both MS/MS experiments performed alternatingly) min: MS/MS 207 and MS/MS ([M+H] + Isoproturon and Diuron; mode more ions : both MS/MS experiments performed alternatingly) Enabling Life Science Tools Based On Mass Spectrometry

2 Results 1. Method Optimization Seven pesticides were chosen from different pesticide groups carbamates: Aldicarb, Methomyl; triazines: Atrazine, Desethylatrazine; phenylureas: Diuron, Chlortoluron, Isoproturon. (Fig. 1). Ionization conditions were optimized with direct infusion experiments, using different solvents and buffer systems. Best response was obtained with a mobile phase of ACN/H 2 O, containing 0.1% HCOOH, under APCI conditions. A rather low APCI temperature of 200 C proved to be best, not only for the thermolabile carbamates but for all compounds. All pesticides are detectable as [M+H] +, with the exception of Aldicarb. Aldicarb is labile under acidic ionization conditions, so that only fragments could be observed (Fig. 2b). In this case the ACN adduct of a specific fragment [2] is observed as most intense ion (m/z = 157). This ion nevertheless can be used as precursor ion for a more specific detection in MS/MS mode, as shown in Fig.2b and 3. For this precursor ion as well as for those of the the other pesticide compounds high conversion efficiencies for the transformation into specific fragment ions were achieved, which is a prerequisite for low detection limits. 3 a) b) Due to the principles of fragmentation in ion trap systems, these efficiencies are in most cases significantly higher compared to triple quadrupole systems ( MS/MS in time vs. MS/MS in space ). Only in the case of small fragments (m/z < 30% of precursor value; phenylureas!) ion trap systems meet their limitations because of the physical laws of ion trapping. However, fragmentation parameters can be adjusted to some extent, so that even in these cases detection in MS/MS mode is possible (Fig. 2d). c) d) Fig. 3: Chromatogram traces and compound mass spectra: a, b) Full Scan mode, c, d) timetable-controlled MS/MS mode. 4 2. Quantitation Procedures and Standards Quantitation was performed with an external standard calibration, using a mix of pure standard compounds. Concentrations covered the range from 0.1 pg/µl to 5 ng/µl, corresponding to absolute amounts of 5 pg to 250 ng on column. One extract of a drinking water and one of a surface water were analyzed as blank samples and additionally spiked with known pesticide amounts. Quantitation was performed in both full scan MS mode (since the matrix content of these samples can be expected to be rather low) and the more specific MS/MS mode. Four of the pesticides nearly coelute under the chromatographic conditions used for quantitation in MS mode, but this situation is not a problem for ion trap systems, since each individual MS scan contains all four signals. Therefore, shapes of all peaks are well defined. In contrast to this, a comparable simultaneous detection with triple quadrupole systems would not be possible, since the system would have to switch between different signals, resulting in a loss of signal and lower duty cycle. For quantitation in MS/MS mode, chromatographic conditions were adjusted to a slightly better separation, resulting in two groups of two coeluting compounds. This situation can be handled by ion trap systems in the alternating MS/MS mode, which means a rapid switching between different MS/MS experiments. 5 capability still maintains proper peak shapes in this case. a) b) Detection limits as low as 0.1pg/µl (5 pg absolute amount on column) for Atrazine were found, together with linear response through the investigated concentration range (Table 1, Fig. 4). Reproducibility was evaluated for the MS/MS mode by using multiple injections of some of the samples (low concentrated standard, medium range standard, spiked samples). RSD values were in the range of a few percent even for the difficult candidates (carbamates), sometimes even better than 1% (Table 1). 3. Water Samples Both blank samples were shown to contain pesticides. In both samples all pesticides could be detected except for Aldicarb (Fig. 6; Table 2). Surface water contained significantly higher concentrations of some of the pesticides, as was to be expected. Fig. 6: Chromatogram traces for a,) blank drinking water, b) spiked drinking water. Quantitation in timetable-controlled MS/MS mode. Concentration differences between blank and spiked samples were compared to theoretical spiking levels and found to be in good consistency (Table 2). While obviously both methods of quantitation full scan mode and MS/MS mode can be used, the more specific MS/MS method yielded better results. 6 Conclusion The results of this work clearly demonstrate the ability of ion trap system to do proper quantitation jobs. An LC-APCI-MS/MS method using an ion trap system for the specific, sensitive and simultaneous quantification of seven pesticides in water samples is shown. Detection limits down to 5 pg abs. amount on column (0.1 pg/µl sample concentration) could be achieved. Calibration curves show excellent correlation coefficients, high linear ranges and good reproducibilities for multiple analyses.
