1 Using the Motic Microscope Cameras Step 1: Going Live! : This mode of the microcope camera allows you to see the images on a computer screen take still photos and even record video in real time. Log in. From the main applications menu, open Class Applications Science Motic Plus Plug camera USB cable into USB port on the computer. On the top menu bar, click the Capture button (camera icon). Now this screen should appear. Adjust camera settings in the panel on the left of the screen. In the Resolution box, select 640x512 from the drop down menu. Move the Exposure slide bar to left until the fps (frames per second) listed in the box to the right is around 60. The picture is probably too dark now, so move the Gain slide bar to right until the picture looks brighter and clearer. Focus the camera by rotating the eyepiece. Uncheck the Enable box for Gamma You may have to play around with the exposure and gain a little to get a good live picture that is not jittery. Step 2: Capturing a picture: Click on the white balance and see if it looks better on, or off. In the live mode that you are now in, click the dark camera icon in the tool bar on the far upper right of the screen. It won t appear as if anything has happened, but a picture has been taken. Minimize this window to get back to the start screen of Motic. Your pictures should be on the right column. You can toggle back and forth between the Motic Image Plus icon and Motic Live Imaging Module. These tabs are at the bottom of the screen. The Live Image Module allows you to take pictures. The Motic Image Plus tab allows you to see all the pictures you have taken. (Continued on back page)

2 You can single click on any picture on the right to see a larger view of it. If you want to delete a picture, move the cursor over the picture, right click and choose close. The picture should now disappear. Step 3: To Save a picture: Select photo from the right column by single clicking on it. Click Save As, NOT SAVE, feature under the File tab. Give it a file name. IMPORTANT: include the magnification that you took the picture with in the file name. For example: Red blood cell 40X. This will be important when you need to measure the specimen Save in: your home folder, specifically in the folder titled My Pictures. In Save as file type box, select jpeg. THIS STEP IS REALLY SUPERDOOPER IMPORTANT! Click Save Step 4: Calibrate the microscope to correct magnification and unit of measurement (microns). This is necessary to do Step 5: Measuring the specimen. Click the icon on your desktop that says Motic Images Plus Click Measure, then Calibration Wizard. Click on the Load Image box. Find the file name, Calibration dot in the (S) School Shared folder under my name. Double click this file. In the field below the Load Image box, drag the cursor over the arrow and select the 10X objective lens. In the field beneath that, drag the cursor arrow over and select the 150 micron choice. Click on the Calibration box at the bottom of the screen. On the next screen click save, then on the next screen click close. NOTE: You must go through this process every time you log into the Motic software. Measrue the calibration dot. It should be 150 microns. (see how to measure in step 5 of this handout) Make sure your measurement is done having selected the 10X lens under the measurement tab.

3 Step 5: Measuring the specimen : NOTE: Specimens cannot be measured in live camera mode. The pictures have to be captured first, then measured in the blue work mode screen. You can also save them in your home directory and do the measurements later. Measuring your specimen Click on File... Open jpeg from your home directory. Click on Measure at the bottom of the screen. Select the lens that was used to take the picture. NOT total magnification. Note: When measuring the calibration dot, make sure you select 10X. Click on Measure icon at top of the screen. Select the measurement you wish to use, such as line. Move the cursor onto the picture and click, hold, drag and release obtain the measurement (see picture above). Click Save to make edits permanent Transfer pictures from your H drive to your flashdrive VERY IMPORTANT NOTE: ONCE YOU SAVE CHANGES TO YOUR JPEG FILE, THEY CAN NOT BE DELETED. YOU CAN MAKE EDITS BY ADDING THINGS AND CLICKING SAVE, BUT ONCE YOU DO THAT IT IS PERMANENT. SO MAKE SURE YOUR CALIBRATIONS ARE CORRECT.

4 Step 6 :Pasting a Picture into Word: Open a Word Document From menu bar Click on Insert. Click on Picture. Find your file by scrolling to the appropriate folder Select the picture you want double click on it or select the word, Insert. Now you can modify your picture by moving around the page, cropping it, drawing arrow to specific structures and labeling pars, adding text boxes, etc. See the instructions below on how to do this. Step 7: To be able to Move your picture in Word: Right click on your picture. Resize it my clicking on the lower right corner and dragging. Right click on the picture. Scroll down to wrap text and then choose Move behind text. Now you can drag your picture around and put it where you choose. Right click on the picture and notice the tab, Picture Tools up at the top of the tool bar in Word.. Click on Insert and Format to choose a variety of ways to modify your picture Step 8: Finally, save your file with a name linking it with the pictures like, Pond Water Lab. Make a hard copy or turn it in digitally to the shared folder. I will tell you which one.
