Description du projet 7 academic, associated partners (from France, Germany, Belgium, Italy, Spain and Lithuania) which are leading researchers from different complementary fields would like to apply for FP7 call. Fluorescence detection of proteins and DNA inside the cells using exogenous probes remains a challenge, and is currently limited to molecular probes. In the present project, the consortium propose to develop innovative nanoparticle-based fluorescent probes presenting multi-color emission that is many-fold brighter compared to molecular probes. This multi-color emission will change in response to interaction with the target biomolecule allowing its quantitative detection with exceptionally high sensitivity, which is particular interest for intracellular detection of peptides and nucleic acids present at extremely low concentrations inside the leaving cells.
2 The consortium plans to develop different types of nanoparticles : organic and silica-based fluorescent nanoparticles and inorganic upconverting nanoparticles. These particles will be further modified with peptides and nucleic acids in order to obtain nanoprobes. Researches will focus on the intracellular detection of nucleic acids (mrna / mirna) and enzymes (Cell Cycle kinases), which are interesting oncogenic markers for cancer diagnostics. To deliver nanoparticles inside the living cells different approaches will be explored, particularly using vector peptides. Systematic characterization of the nanoparticles by the consortium will address a number of fundamental problems: nanoparticle photophysics, stability and photostability, the biosensing mechanisms, toxicity and phototoxicity as well as nanoparticle delivery inside the cells, etc. A broad variety of fluorescence techniques be applied: multi-color fluorescence microscopy, fluorescence lifetime imaging, single molecule detection, single-particle tracking, high content arrayscan cellular imaging, combined fluorescence and AFM, etc. As the project is highly interdisciplinary, dealing with nanosciences, photophysics, bioconjugate chemistry, cell biology variety of instrumental techniques for bioimaging, the consortium will provide broad range of training of PhD students through research.
