It encodes a nuclear p53 protein of 393 amino acids, which responds to diverse cellular stresses to regulate target genes that induce cell cycle arrest, DNA repair, apoptosis, senescence or changes in metabolism. Tumour-associated alterations in TP53 are a hallmark of over 50% of human cancers and cause dramatic defects in p53 function promoting the accumulation of DNA damage. MDM2, a p53-inducible phosphoprotein, binds to the N-terminus of the p53 and negatively regulates its transcriptional activity.
