3 4. Scientific Activity 4.1. Scientific Training 2003 2004 "Adhesion of Desulfovibrio alaskensis to glass surfaces" conducted in the Biocorrosion laboratory at the Faculty of Science, University of Lisbon under the guidance of Professor Maria Margarida Meireles Teixeira de Faria (Faculty of Science University of Lisbon) and Dr. Paula Bacelar-Nicolau (Open University), September 2003 to October 2004, with final score of 18 points (1 to 20). This traineeship (one year) consisted in learning microbiology techniques, in anaerobic conditions, and routine light microscopy. 2007 Traineeship in the laboratory of Professor Harry Gilbert at the Institute for Cell and Molecular Biosciences, Medical School, Newcastle University. This traineeship (3 months) aim was to obtain calorimetric values of the formation of protein-protein complexes. It was learned the handling of the isothermal calorimetry titration apparatus (ITC). 2008 Traineeship (3 months) in the laboratory of Professor Harry Gilbert at the Institute for Cell and Molecular Biosciences, Medical School, Newcastle University. The main goal of this traineeship was to consolidate the calorimetric data of protein complexes under study. 2009 Traineeship (3 months) in the laboratory of Professor Harry Gilbert at the Institute for Cell and Molecular Biosciences, Medical School, Newcastle University. 4.2. Research Area Study in Protein Biochemistry. Employment of molecular biology techniques such as polymerase chain reaction (PCR) and cloning into plasmid for protein expression. Employment of various techniques of protein purification, including chromatography on immobilized metal affinity (IMAC), size exclusion chromatography and polyacrylamide gel electrophoresis (native and denaturing). Application of techniques of crystallography as protein crystallization techniques (suspending drop, sitting drop, crystal seeding), crystal fishing and freezing, synchrotron data collection and analysis, model building and phase refinement. Utilization of isothermal titration calorimetry for obtaining calorimetric parameters of protein-protein binding. Investigation of proteins acting on carbohydrates and complex enzymatic degrading of plant cell wall. 4.3. Research Activity 3

The principal aim of the attendance of this course was the introduction on the use of bioinformatics tools for the analysis of protein and nucleotide sequences. Participation in the course "Protein Crystallography", Department of Chemistry, Center of Fine Chemistry and Biotechnology, Faculty of Science and Technology, New University of Lisbon (26 to 30 March 2007). It allowed the initiation on the methodology used in obtaining and studying protein crystals and thereby obtaining structural models of proteins. 4
