ZORBAX HPLC Columns The Agilent ZORBAX columns C8, C8, and Phenyl are designed to provide good peak shape for basic, acidic, and neutral compounds over the ph range -9. This wide useable ph range allows superior method development flexibility with one family of columns. columns can be used for initial method development at low ph (-) and will provide high resolution and excellent peak shape of all types of compounds. The same column can be used for method development in the mid ph (6-8) region. In the mid ph region, residual silanols are more active and tailing interactions are more likely. To overcome these interactions, columns are extra Densely Bonded and double endcapped through a proprietary process to cover as many active silanols at possible. The result is superior peak shape of basic compounds from ph -9. columns are available in C8, C8, and Phenyl bonded phases for selectivity optimization of most separations. StableBond (for low ph), Bonus-RP (for low and mid ph), and Extend-C8 (from low to high ph) columns are available as additional selectivity choices. Extra Densely Bonded and Double Endcapped Bonded Phase A Good Peak Shape over a Wide ph Range with ZORBAX ph Excellent first choice for method development Excellent peak shape for bases, acids, and neutrals Wide useable ph range ph -9 High purity silica Long lifetime with extra dense bonding and double endcapping ZORBAX columns provide good peak shape over a wide ph range and are an excellent first choice for method development from ph -9. ZORBAX HPLC Columns Column Stability Testing at ph and 60 C K of Amitriptyline ZORBAX SB-C8 P/N 88975-906 ZORBAX -C8 P/N 99967-906 Competitive C8 Column Volumes of Mobile Phase (X000) Column Stability Testing at ph 7.0 Competitive C8 SIL-type After 86 Column Volumes Column: 4.6 x 50 mm, 5 µm Purge Conditions: Mobile Phase: 70% 50 mm NaAc-HCl, ph.0 0% ACN Retention Test Conditions Mobile Phase: 65% Methanol 5% Water Flow Rate:.0 ml/min Temperature: 60 C Sample: Tricyclic Antidepressants columns are stable over a wide ph range. At low ph, Eclipse, an end capped column is extremely stable and shows equivalent stability to a non-endcapped column, SB-C8, at ph. The columns were purged with a ph mobile phase at 60 C. Then they were tested with a strongly basic compound to determine if the endcapping or bonded phase had been hydrolyzed from the silica surface. The column was very stable, as shown by the consistency of the retention of amitriptyline over the,000 column volumes of the test. Another endcapped column shows less stability under these same conditions. Double endcapping, dense bonding and the durable Rx-Sil particles (sol-type) combine to provide long lifetime at ph 7 when compared to single endcapped sil-gel columns used here. The conditions used for this test high temperature (60 C) and high salt concentration (50 mm), accelerate the dissolution of silica, causing premature failure of the sil-gel type column. -C8 P/N 99967-906 Sol-type After 84 Column Volumes Column: 4.6 x 50 mm, 5 µm Mobile Phase: 60% ACN 40% 50 mm Phosphate Buffer, ph 7.0 Flow Rate:.5 ml/min Temperature: 60 C Sample: Tricyclic Antidepressants. ZORBAX HPLC Columns Selectivity Changes for Basic Compounds with and Stable Bond Double Endcapped -C8 P/N 966967-906 Keep your Agilent 00 Series HPLC system running at peak performance To ensure that your LC pump, manual injection valves, autosampler, and detectors work at maximum efficiency, request a copy of Maintaining your Agilent 00 Series HPLC System. This 55-page resource will help you maximize uptime and increase productivity. Barbital. Sulfamethoxazole. Caffeine and StableBond columns are based on the same silica but have different bonding and endcapping. Therefore, they can have very different selectivity for the same sample under the same conditions, as this example shows. -C8 P/N 96967-906 -C8 8 P/N 96967-90 0 4 6 8 0 4 6 Column: 4.6 x 50 mm,.5 µm Mobile Phase: 5% H O: 85% MeOH Flow Rate:.0 ml/min Temperature: 0 C Detection: UV 0 nm Sample:. Oxybenzone. Padimate-O. Ethylhexylsalicylate min This separation of sunscreens on all three bonded phases C8, C8 and Phenyl shows that different bonded phases can be used to optimize a separation. While all three bonded phases provide an adequate separation, the -Phenyl provides a different peak elution order and a much shorter overall analysis time. All three bonded phases also provide excellent peak shape with no mobile phase additives.
