FUEL ETHANOL APPLICATION SHEET Novozymes Spirizyme products for use in saccharification and fermentation The Spirizyme product range offers a wide variety of possibilities to optimize yields, throughput, and process consistency. Spirizyme products are the leading industry choice in plant start-ups and established operations. Spirizyme products maximize starch and dextrin conversion to sugars for yeast fermentation to ethanol. Benefits Saccharification enzymes are used for the extraction of fermentable sugars from starch in saccharification and fermentation processes. Ethanol plant performance is influenced by raw material quality and process variation. Products Spirizyme Excel is the most advanced saccharification solution, designed to convert more of the difficult starch. For use in plants seeking maximum value extraction and robust performance for variable mash and process conditions. The product is highly concentrated and contains a unique composition of glucoamylase, alpha-amylase, and debranching activities. Spirizyme Ultra is a premium glucoamylase product which provides consistently high ethanol yields for plants focused on high-value extraction. It is a highly efficient and robust glucoamylase, resulting in low levels of residuals. Spirizyme Ultra is a highly concentrated product. Spirizyme Fuel HS is the industry standard glucoamylase for plants whose primary concern is low enzyme use cost. Use of this product provides high ethanol yields and the fastest glucose generation available with the best thermostability. Performance Spirizyme glucoamylases are used on liquefied starchcontaining substrates to produce sugars for fermentation. The enzyme preparations work in dedicated saccharification stages as well as simultaneous saccharification and fermentation (SSF) operations. The glucoamylases hydrolyze both 1,4- and 1,6- alpha linkages to liberate glucose for subsequent fermentation by the yeast. Products differ in specificity and action pattern, allowing for options in customizing the use for specific plant operations. The activity performance of Spirizyme products relative to ph conditions is shown in Figure 1. Fig. 1. Performance on corn-based mashes The fermentation performance (SSF) of Spirizyme products is shown in Figure 2. Fig. 2. Fermentation rate in general depends on mash characteristics, enzyme type, and dosage strategy. The ability to achieve higher conversion of starch without causing yeast stress influences both ethanol yield and time to complete fermentation. Figure 2 shows the fermentation profiles obtained when using industry average dosages of Spirizyme Fuel HS, Spirizyme Ultra, and Spirizyme Excel. Figure 3 shows the relative ethanol yield performance of the three products when fermenting the same mash. It can be seen that the three products provide significant difference in ethanol yield potential for this mash. Page 4 of 6

Performance of glucoamylase products may vary according to conditions specific to each ethanol production plant. The optimal product choice may vary according to plant conditions and objectives and should be discussed with Novozymes representatives. Guidelines for conducting evaluation trials are available. Please consult your Novozymes representative for a more detailed recommendation to serve your specific needs. Performance on other feedstocks The optimal choice of glucoamylase for other feedstocks than corn will vary. Contact your Customer Solutions representative to find out what is the best choice for your feedstock (e.g., wheat, barley, cassava, sorghum). Page 5 of 6

Usage Dosage Table 2 describes typical dosing scenarios for Spirizyme products. Note: Optimal dosages must be determined under actual plant operating conditions. Storage in application Spirizyme products should be stored at cool temperatures in closed containers protected from sunlight. The products have been manufactured for optimal stability; however, enzymes gradually lose activity over time. Extended storage and/or adverse conditions such as high temperatures or increased humidity may lead to a higher dosage requirement. Contact your Novozymes representative for detailed storage recommendations and cleaning procedures relevant to your facility.
