Rapid DNA Analysis in the Police Booking Suite: FBI Initiative for Reference Sample Point-of-Collection Analysis 13 th European Forensic DNA Working Group Meeting Krakow, Poland May 10, 2012 Clark Jaw - FBI Laboratory

Overview 1. Rapid DNA Definition 2. SWGDAM R-DNA Activity 3. R-DNA Test Plan Collaboration 4. Advisory Policy Board (APB) 5. R-DNA Task Force (R-DNA TF) 6. R-DNA Integration Phases

Rapid DNA Typing Systems Develop a fully integrated system capable of performing STR analysis of buccal swabs in 1-2 hours, independent of a DNA laboratory. Goals: Little user interaction (or experience) Rugged Robust Automatic data interpretation 4-8 (or more) samples per run Disposable chips (with reagents on board) Swab In Answer Out

Testing and Validation of a Rapid DNA System Purpose of the instrument: Generate a STR profile for each reference sample buccal swab analyzed in the instrument Rapid DNA Instrument The Box (Various vendors) Common message format File for CODIS Text File A swab is collected from an individual Swab is loaded into the instrument A disposable cartridge containing reagents for DNA extraction, PCR, and separation is loaded Push go The basic workflow 1-3 hours The electropherogram is interpreted by an (integrated) expert system software The appropriate genotypes for the sample are collected

Vision of R-DNA Integration (Non-Laboratory Analysis) SWGDAM Industry R & D Laboratory FBI/NIST/LE Laboratory Performance Testing State Offender Concordance Validation SDIS State/Local Crime Scene Laboratory Pilots Police Booking Station DNA Task Force- APB

NDIS Rapid DNA Typing Systems Under Development Systems for buccal swab analysis are currently under development and are not yet commercially available These are STR-based instruments running the genetic marker systems currently used by law enforcement (CODIS-FBI NDIS)

SWGDAM R-DNA Committee Ad Hoc met three times from September January 2011 Recommendation to SWGDAM Chair to establish full Committee January 2011: R-DNA Committee established Goals: Evaluate/test prototype of R-DNA instrumentation Establish requirements and timeline for pilot studies Committee Memebers: Taylor Scott (IL State Police) Chris Carney (FDLE) Rhonda Criag (FBI ndna Unit) Theresa Caragine (NYC OCME) Pete Vallone (NIST) Elizabeth Johnson (USACIL DOD) Julie French (MI State Police) Lilliana Moreno (FBI ndna Unit) Clark Jaw (FBI CODIS Unit) Richard Guerrieri (FBI FDDU)

SWGDAM R-DNA Committee July 2011 First Meeting Performance and Test Plan discussion (NIST) Validation requirements discussion What is necessary for a Pilot study (est. threshold for data use: Controls, QA, Validation, etc.)

SWGDAM R-DNA Committee Initial discussion of a universal file format for electropherogram data (fsa, HID) In addition to R-DNA data ( allele calls ) the ability to export the e-gram file in a universal format CODIS Committee supported universal e-gram format How to proceed/discussion?

SWGDAM R-DNA Committee Positive Controls? Run with every run, once a week/month/year? Effect on sample throughput and cost example: a 4 channel system (25% of analysis) Evaluation experiments will help guide Cell-line DNA on a swab, operator buccal swab, other, non-human spike (internal control)?

SWGDAM R-DNA Committee Can not go forward with database pilot studies until performance evaluation and developmental validation are completed Laboratory pilot studies: ~ 2-3 years Booking Station: ~ 3-6 years

SWGDAM R-DNA Committee Initial discussions of validation Results of initial evaluation testing will help determine what is required for performance metrics Concordance high priority Threshold for failure reliability Developmental validation Performed by vendors, testing groups NDIS Acceptance Validation Benchmark Internal validation Concordance (# of samples?) Sensitivity Sizing precision, resolution Certification Can these instruments be certified? These discussions to follow Performance evaluations

R-DNA Instrument Test Plan Collaborations NIST Human Identification Group Technical assistance in assessment of development efforts Test Plan development Testing involvement for RapidHIT 200 and ANDE (Allele calls and e-grams) NetBio ANDE Test Plan NIST set of ~ 80 numbered samples with protocol for analysis USACIL, DHS, FBI and NetBio IntegenX RapidHIT 200 Test Plan NIST set of ~80 numbered samples with protocol for analysis DHS, FBI and IntegenX Performance will directly feed into second generation development FBI will re-evaluate decision to procure and test only integrated machines

SWGDAM R-DNA Committee Booking Station Issues (3-6 years) Sample/cartridge disposal chemical waste Queuing of samples If booking station throughput is low How will validation be addressed? Can the instrument be certified? Will a performance check of a certified instrument be acceptable?

APB R-DNA Task Force Advisory Policy Board (APB) Federal Advisory Board for FBI Criminal Justice Information Services Agiro Magers - Maryland State Police James Buckley - DHS Immigration and Customs Enforcement (Federal WG) James Gerst FBI Criminal Justice Information Services Division Dave Hober - Assistant Chief, San Jose Police Department Mike Lesko - Texas DPS Chairman Identification Service Subcommittee James Loudermilk - FBI Science and Technology Branch Terry Maketa - Sheriff, El Paso County, Colorado William Reed - Virginia State Police Sherry Sabol - FBI Office of General Council Diane Urban - Chief, Hayward CA Police Department David Coffman - Florida Department of Law Enforcement Ted Staples - Georgia Bureau of Investigation Julie French - Michigan State Police Hope Olsen - North Dakota, Office of Attorney General Crime Laboratory Tom Callaghan - FBI
