Duing this taining, ou enginee will guide you though the basic opeation of the system, eview safety and maintenance guidelines, and eview optional accessoies that povide optimum pefomance fo you system. The pupose of basic taining is to intoduce you to the system and ensue that you ae able to pefom basic functions. Howeve, the taining is not intended to include ceating custom applications. Fo advanced taining options, please contact you sales office. The following peequisites have been defined to ensue a successful basic taining: 1. The opeato must be available without inteuption fo the entie taining session. 2. No moe than two opeatos will be tained as pat of the installation. The system opeation equies a basic knowledge of Windows XP. Ceate a new method fo a constant voltage sepaation in a 60 cm, 75 µm I.D. capillay. 2. Pepae the following 2 ml vials fo inlet and outlet vial positions and fo inses: Vial Contents Volume Position Rinse Run Buffe A 2 ml BI:B1 Injection Run Buffe A 1 ml BO:B1 Sepaation Run Buffe A 2 ml BI:A1 Sepaation Run Buffe A 2 ml BO:A1 Waste N/A 0 BO:A3 S = Sample; B = Buffe I = Inlet; O = Outlet A1 though F6 indicate Vial Positions 3. Fo UV and PDA detectos, fill one 2 ml vial with Beckman Coulte Test Mix B and place in the SI:A1 position. If you ae using an LIF detecto, fill the 2 ml vial with Beckman Coulte LIF Detecto Test Mix. (Pepae LIF Detecto Test Mix accoding to the Test Mix diections.) 4. Set the initial tempeatue to 23 C. 5. Pogam an un-timed pe-inse (with a duation of at least two minutes at psi) of Run Buffe A (BI:B1) to the waste vial (BO:A3). 6. Pogam a 10 second fowad pessue injection of Beckman Coulte Test Mix (SI:A1) at 0.5 psi to the injection buffe vial (BO:B1). 7. Pogam a 30.0 kv constant voltage sepaation using a 0.17 minute amp time and a duation of 6.0 minutes. Select the At Time check box and ente 0.00 minutes. 8. If you ae using a UV detecto, set the wavelength to 214 nm. If you ae using a PDA, set the wavelength to 214 nm with a 10 nm bandwidth.
