2 A checklist for designing an animal facility what should you ask and who should answer?

3 The design team The planning and building of a laboratory animal facility is a complex achievement involving the participation of expertise in several fields and the first step is to set up a design team or committee consisting of: Representatives of the client: Head of the planned facility User representatives Animal technicians Administration representatives People responsible for the future maintenance of the building Professional team Architects Engineers Consultants?????

4 The next step would be to define activity Type of research activities: Development (toxicology, product testing etc) or Discovery (basic research, applied research etc.) Or Regulatory control GLP or non-glp?

5 Species identifying the variety of species expected to be accommodated over time: Mammals: rodents, lagomorphs, large animals (dogs, pigs), primates Aquatic species: marine, fresh water Insects

6 Containment - risk of contamination to researchers and/or the outside environment? Infections Chemicals Isotopes There are well described European and national laws that regulate such usage. Should there be a need to work with such agents it is essential that a special working group be a part of the design team to source and implement these rules. 7 Sources of animals In-house breeding Commercial sources Non-commercial (academic institutions, wild caught animals) These questions should be asked and answered before starting on trying to get an estimate of capacity.

8 Capacity: Strategic limits and questions A large laboratory animal facility is somewhat like a hotel when planning capacity. Planning needs to be based on the average number of housing units per day that is balanced against the total number of animals that can be expected to be housed in the course of any given research period. It is also useful to get an idea of long term trends in the research environment that will use the facility. An example is whether the facility will use large and small species at any time in the future. Exclusion of large species is a major strategic decision and will influence all planning going forwards. The animal facility should be designed with high degree of flexibility and expansion possibilities. This is particularly the case when designing for discovery research.

9 Questions to ask: Rodents and/or large animals only Special species with specialised environmental requirements (nude mice or rats that require 2 to 3 degrees higher than normal small rodents) Primates yes/no Work with pathogens (human or animal) yes/no Separate building or integrated into research building All in-vivo laboratories included within the facility (behind the barrier?) or not In-house breeding or not (stringent barrier requirements)

10 Barriers Isolation, Defined as to keep out or to provide a barrier and containment, or Defined to keep in or to provide an envelope around an experiment or animal room, are provided in response to the specific requirements, and may have to be met at the same time. As there is no such thing as a perfect barrier, the design and planning process of a laboratory animal facility consists of sorting out priorities and establishing compromises (Ruys).

11 BARRIERS There are such a vast amount of evidence that clearly states that intercurrent disease has a negative effect on biomedical research that we cannot longer accept research in completely open uncontrolled systems. All modern animal facilities should be planned with varying degrees of barrier confinement. The barrier concept is designed to provide a stable environment that ensure reproducible research. Its is also designed to keep undesirable agents (infections environmental) out of the facility. The barrier will also contribute to reducing exposure to allergens as well chemical and physical hazards that may be used within research.
