Incubate at 41.5±1 C during 24±2 hours Malaquite green inhibits the growth of accompanying flora Magnesium cloride allows a better growth of Salmonella After incubation there is a lot of Salmonella cells

5 Buffered Peptone Water Peptone / Salts / Buffer Calcium carbonate / Tetrationate / Ox bile / Brilliant Green / Novobiocine 1 ml of the incubated buffered peptone water is transfered into 10 ml tube of. Incubate at 37±1 C during 24±2 hours Tetrationate inhibits the growth of intestinal bacteria Brilliant green prevents the growth of Gram negative bacteria Bile stimulate Salmonella growing and inhibits accompanying flora Novobiocine inhibits the accompanying flora After incubation there are a lot of Salmonella cells

6 Buffered Peptone Water Cat Chromogenic Cat.1122 XLD Agar (Xilose Lisine Desoxicholate) Peptones / Salts / Buffer Xilose, Lactose, Sucrose & Phenol Red / Sodium Thiosulfate y Ferric Ammonium Citrate / L-lysine Incubate at 37±1 C during 24±3 hours If Xilose, lactose and sucrose are fermented, the ph decrease, changing the color of phenol red, that is a ph indicator Sodium Thiosulfate provides Sulphur and Ferric ammonium citrate is the indicator for H 2 S production Lisine is decarboxilated, producing cadaverine, which increases the ph and forming a purple halo around the colony Salmonella growth in black centered red colonies

7 Buffered Peptone water SALMONELLA CHROMOGENIC AGAR Peptones / Salts / Chromogenic Substrate Incubate at 35±1 C during hours Esterase activity cleaves one of the chromogenic substrates, giving the colony a magenta color β-galactosidase activity cleaves the other chromogenic substrate, giving the colony a green color Salmonella growth in magenta colonies

8 Buffered Peptone water NUTRIENT AGAR Peptone / Salts / Buffer Incubate at 35±1 C during hours Medium rich in nutrients Isolated colonies for biochemical tests

9 Buffered Peptone water TSI AGAR Salt / Buffer Glucose / Lactose & Sucrose Phenol Red / Ferric ammonium citrate & Sodium thiosulfate Incubate at 35±2 C during hours Glucose fermentation turns the medium into yellow and may product gas Lactose and Sucrose fermentation turns the medium into red Sodium Thiosulfate provides Sulphur and Ferric Ammonium citrate, which is the indicator for H 2 S production

10 Buffered Peptone water LYSINE DECARBOXYLASE BROTH Glucose / Bromocresol Purple / Lisine Incubate at 37±1 C during 24 ± 3hours Glucose fermentation turns the medium into yellow Lisine is decarboxilated, producing cadaverine, which increases the ph and forms a purple halo around the colony Tubes with a positive reaction to lisine decarboxilation will present a purple color
