3 I giorni 19 e 20 dicembre 2013 alle ore 9,00, presso la Stanza A325 del Ministero della Salute, sito in Roma in Via Giorgio Ribotta, 5, si è riunito il Comitato di Valutazione per i progetti giovani ricercatori, afferenti al Bando R.F , nella composizione di cui al D.M. 4 dicembre Partecipano i componenti di seguito riportati: Dott. (All.1). Il Dott. Massimo Casciello, Direttore Generale della Ricerca Sanitaria e Biomedica e della Vigilanza sugli Enti, porge il benvenuto ai presenti e comunica che la conduzione della study session sarà affidata alla Dott.ssa Chiara Gabbi in qualità di Presidente del Comitato di che trattasi, al Dott. Gaetano Gugliemi, direttore dell Ufficio III in qualità di referente del processo di valutazione e alla Dott.ssa Maria Novella Luciani, Direttore dell Ufficio IV, referente per i dettagli organizzativi e per i rapporti nella fase audit con la Commissione Nazionale per la Ricerca Sanitaria. Gugliemi, nel salutare i componenti specifica le procedure di assegnazione e di valutazione effettuate dai referees per i progetti Giovani Ricercatori presentati nel Bando Ricerca Finalizzata , puntualizzando che i progetti presentati in questa sezione sono in totale 1218 (All. n. 2) di cui 3 con una sola valutazione e 283 con due valutazioni ma senza F2F (face to face) concluso. Comunica, preliminarmente, le procedure da seguire nel corso della study session e, presentata ai componenti la situazione del referaggio effettuato, espone le modalità di accesso al sistema, per le quali sono attribuite a ciascun membro le proprie credenziali informatiche. Rappresenta, inoltre, che per la prima volta è stato attivato un sistema di Editors che ha controllato la corrispondenza dei punteggi dati, in rispetto alle linee guida e che eventuali non corrispondenze saranno indicate alla study session attraverso una specifica segnalazione nel cruscotto di valutazione. 2

4 La Dott.ssa Chiara Gabbi dà avvio alla discussione per la definizione collegiale dei criteri da seguire nel corso dei lavori. Il Comitato prende quindi atto che i progetti - GR ; - GR ; - GR ; - GR ; - GR sono stati posti in valutazione in via cautelativa e che la decisione finale sul rispetto dei requisiti amministrativi previsti dal Bando è di competenza della Commissione Nazionale Ricerca Sanitaria. Il Comitato assegna, poi, i seguenti progetti con una sola valutazione, affinché al termine possa risultare che tutti i progetti abbiano due valutazioni individuali e indipendenti: - GR ; - GR ; - GR I commissari, dopo aver esaminato i progetti con un solo giudizio, avviano un ampia discussione collegiale al fine di condividere il punteggio finale da assegnare. Il Comitato, successivamente, passa all esame dei progetti primariamente con due valutazioni senza face to face concluso (All. n. 3). Il Comitato decide, altresì, di rivedere tra tali progetti con particolare attenzione quelli con entrambi i giudizi con un delta superiore a 5. Alle ore 12 del 19 dicembre 2013, il Ministro porge i saluti e augura buon lavoro ai partecipanti, ringraziandoli della fattiva collaborazione e sottolineando l importanza e l eccellenza dell attività di ricerca svolta anche all estero dai giovani ricercatori italiani. I lavori vengono sospesi alle ore 19,00 del 19 dicembre 2013 e riprendono alle ore 09,00 del 20 dicembre Il Comitato procede quindi alla rivalutazione di n. 6 progetti il cui giudizio del face to face risulta sospeso perché rifiutato da uno dei due referee. 5 Concluse tutte le fasi di valutazione viene visualizzata la situazione completa di tutti i progetti. Dopo ulteriore verifica della coerenza del giudizio espresso dai referee sia nelle valutazioni individuali che nel face to face con il punteggio attribuito, il Comitato di Valutazione Giovani Ricercatori collegialmente procede alla stesura dei relativi commenti di valutazione (All. n. 4) e alla stesura della graduatoria finale, approvandola, come da elenco allegato al presente verbale come parte integrante dello stesso (All. n. 5). Il Comitato suggerisce quindi, tenuto conto dell elevata qualità scientifica dei progetti presentati, di proporre alla Commissione Nazionale per la Ricerca Sanitaria di assicurare il finanziamento di tutti i progetti con un final score pari a 12 eventualmente anche attraverso una riduzione economica che, comunque, non infici la loro realizzazione. 6 BANDO RICERCA FINALIZZATA-GIOVANI RICERCATORI COMITATO VALUTAZIONE GIOVANI RICERCATORI Elenco Motivazioni GR IDENTIFICATION OF NEW GENES FOR RARE NEURODEVELOPMENTAL DISORDERS BY HOMOZIGOSITY MAPPING/NEXT GENERATION SEQUENCING AND FUNCTIONAL STUDIES IN ZEBRAFISH This project focuses on use of a multimethod approach to discover novel causative genes in neurodevelopmental disorders. The topic is relevant to National Health System and appears feasible. The methodology includes homozigosity mapping, NGS and Zebrafish model. Beside the limited originality to use NGS to identify novel responsible genes, the impact of the outcome make the study a potentially important project. The rationale is well defined, the facilities and the budget are adequate. Strenghts include the preliminary data, focusing on a similar exploratory-study, and the adequate profile of the PI and his team/collaborators (UO1 and UO2). Weakness include the poor information about the specific selection of families to be enrolled in the study. The impact of the identification of novel genes responsible of neurodevelopmental disorders could be important for a better diagnosis and therapeutic approach in the future. After reviewing the scores and the comments of both the reviewers, the committe agree with the score of reviewer 2 and agree with a final score of 10. Final Score: 10 GR Role of type I IFN in dictating the immunogenicity and therapeutic efficacy of chemotherapy: from animal models to humans The objective of the present proposal is to investigate whether the immunogenic potential and, consequently, the therapeutic success of defined chemotherapeutic drugs resides in the early induction of type I IFN within tumor bed and to elucidate the underlying molecular mechanisms. The PI has a very good publication track record and exceptional collaborators. Istituto Superiore di Sanità is an outstanding environment for the proposed studies. Budget is appropriate. The proposal is of high scientific quality. The team members are appropriate in terms of availability and complementarities of all the relevant expertise, and in how the different roles and responsibilities are distinguished. The applicant is well established in the field. Budget is adequate. The scores assigned by the reviewer #1 do not match the comments. Many criticisms have been noticed, mainly regarding preliminary dataand methodologies. The overall hypothesis is not clear, affecting the scientific quality of the study. Therefore, the committee has decided to validate the average between the scores. Delta score is high because both reviewers have used high scores, making the difference to look higher. After reviewing the proposal and the comments of the reviewers, the committee confirms the average between the two comments. Final Score: 27,5 GR Unraveling the genetic complexity of myeloproliferative neoplasms and the molecular pathways involved in disease progression The present proposal aims to perform whole-exome sequencing to identify mutations in Jack-, Mpl-, normal karyotype MPN samples. The studies may lead to the identification of novel mutations that contribute to progression to AML. Overall the proposal has been well written. Nevertheless, two major concerns have been raised. First, it is not clear whether the Applicant will be able to reach a significant number of samples that may support any statistic significance. Second, the idea of performing whole exome sequencing may be a limitating factor, since putative relevant mutations may occur in noncoding regions. The PI responsible for the submitted proposal has expertise in the specific field, as documented by the publication in medium-impact factor Journals. The Institution is adequate for completing the proposed studies. The Committee has agreed on a final score of 13.5, as in the proposed F2F score. Final Score: 13,5 GR BASAL GANGLIA FUNCTIONAL ABNORMALITIES CORRELATING WITH GAIT DISTURBANCES IN PARKINSON'S DISEASE. TOWARD NOVEL BRAIN SIGNAL CONTROLLED DEVICES FOR ADAPTIVE DEEP BRAIN STIMULATION. The present study aims to develop deep brain stimulation (DBS) patterns to prevent and inhibit freezing of gait (FOG), which is one of the major disturbances in Parkinson s Diseases. The applicant should clarify what is meant by adapting DBS to moment to moment clinical states. If successful, the proposal could eventually contribute to improvements in the efficacy of DBS in reducing freezing of gait and improving outcomes in Parkinson's disease patients. The proposal is well written and has logical aims. The novelty of the study relies in the development of a system that is responsive to freezing of gait. Facilities and infrastructure are appropriate for the proposed studies. Budget is also appropriate. The applicant is a young investigator at the first year of the PhD training. She is first author in several publications related to the topic of the proposal. Complementary expertise will be provided by the assembled team. Despite the high potential of the candidate to 720 8

14 develop an independent career, this reviewer has concerns regarding the leadership of the candidate and the feasibility of the proposed study. Applicant has a good CV and publication records but not the same for co-workers. The committee proposes to accept the evaluation of the second reviewer. 17 GR Development of an engineered human ipscs derived bio pacemaker as an in vitro platform for drug screening This is a potentially interesting proposal that aims to verify the feasibility of in vitro generating human ipsc-derived engineered bio-pacemakers and their suitability as platforms for drug screening in order to test cardiotoxicity or proarrhythmic actions. Nevertheless, the overall interest is lowered by the lack of clarity in regards to the working methodologies and experimental procedures that appears to be not well focused, nor well described. The PI responsible for the submitted proposal is a Young Investigator that has shown proven to be a promising researcher as documented by the relative track of publications listed within the application. The Committee has agreed on a final score of Final Score: 17,5 GR Hospitalization and comorbidities in patients with parkinsonism and / or dementia: a study of prevalence and management proposals scores. Final Score: 22 GR Guidelines for the identification of quality standards for the realization of extra hospital structures for subjects in Vegetative or Minimally Conscious State The two Reviewers were concordant about the limitations of the publication background of the PI. The possibility to recruit a sufficient number of cases to run a reliable analysis for the construction of national guidelines remains questionable. After reading the scores and the comments of the Reviewers, the Committee accepts the proposed score issued from the face-to-face agreement of 23. Final Score: 23 GR Genetic basis of steroid resistant nephrotic syndrome and implications for therapy Status F2F: Rejected After reviewing the scores and the comments of the reviewers, the committe agree with the average final score of 12,5. The expertise of the applicant in the field is very good but the publications record (excellent for one paper) is limited. The project is ambitious and it is not well hypothesis driven, as indicated by the reviewer 2. Final Score: 12,

Final Score: 9,75 GR Mineral metabolism and microvascular function in patients with chronic kidney disease This proposal aims to investigate the role of 1,25dihydroxyvitamin D (1,25(OH)D) and Parathyroid Hormone in the pathogenesis of cardiovascular events in patients with chronic kidney insufficiency. Although this project has interesting ideas, the study design is extremely weak. For instance, patients have to be stratified according to the different etiologies of the renal disease as well as to other independent cardiovascular risk factors. Therefore a sample size of 20 appears to be insufficient to reach a significant statistical power. PI is MD, resident in nuclear medicine, with only 1 paper in the same field of the proposal. No senior authorships, raising the concern on the effective leaderships in this proposal. 22 GR Toward identification of biomarkers in patients with irritable bowel syndrome (IBS) scores. Final Score: 19,75 GR Can default and attentional network dysfunctions lead to Visual Hallucinations and Delusions in Lewy body disease? This project aims to investigate the aetiology of visual hallucinations (VH) in Lewy body diseases (LBD) using an original combined MRI/fMRI/DTI/MRS methodology. The proposal was evaluated by two reviewers. Both reviewers found that the proposal presents good resources and facilities. They also found the project innovative and well written. One of the reviewer found in the proposal some weaknesses that include the unclear role of the units for some aim and the uncertainity of the therapeutic outcome. The committee agrees that these weaknesses limit the impact of the proposal and also notes that the proposal might benefit from more experienced collaborators, more articles as first-author of the applicant and a clearer evaluation of the study design in the proposal. For the above reasons the committee suggests a final score of Final Score: 14 GR Association between an increased systemic inflamation level in periodontitis and cardiovascular diseases: a randomized protocol.
