Mycoplasm oplasma elimination with Mynox Gold: Application in detail Biochrom AG Information Mynox Gold represents the further development of the classical Mynox, which was known for a very high elimination efficiency and low cytotoxicity Mynox Gold is a combination of a standard antibiotic and a biological reagent. One application comprises of 4 vials, a starter treatment and three main treatments. The starter treatment kills most of the mycoplasma particles without harming the cells. The main treatment kills all remaining mycoplasma leading to a permanent eradication with efficiency rates up to 100 %. One application can be used to cure one cell culture permanently from mycoplasma burden. 1 Application and test principle Contamination of cell cultures by mycoplasma occurs frequently. For both biological and economical reasons, it is important to eliminate mycoplasma from cell cultures being used for basic research, diagnostics, and biotechnological production. The most commonly used method for elimination, inactivation, or suppression of mycoplasma in cell cultures is treatment with antibiotics. In general, antibiotic therapies alone do not result in long-lasting, successful elimination. Also, the cytotoxic properties of antibiotics can cause undesirable side effects on eukaryotic cells and can facilitate the development of resistant mycoplasma strains. Mynox Gold (cat. no. W 10-0201/0501/1001) represents the further development of the classical Mynox. Mynox Gold is a combination of a standard antibiotic and a biological reagent. In comparison to mammalian cells, mycoplasma lack a cell wall but are encircled by a cytoplasmic membrane. This biological reagent integrates into the mycoplasma membrane and compromises its integrity. By the combination with a standard antibiotic, the effective dose of both, the reagent and the antibiotic, can be reduced to a minimum for lowest cytotoxicity, still causing a highly reliable and definite elimination of mycoplasma. These biophysical properties making the development of resistant strains highly unlikely. One application comprises of 4 vials, a starter treatment and three main treatments. The starter treatment kills most of the mycoplasma particles without harming the cells. The main treatment kills all remaining particles leading to a permanent eradication. Mynox Gold is intended for research use only. 2 Reagents and materials 2.1 Kit components Each pack contains 1 vial starter treatment and 3 vials main treatment. 2.2 Stability and storage Mynox Gold is stable until the expiry date given in the bag label if stored at +2 C to +8 C in the dark. Shipping at room temperature. 2.3 Supplemental requirements standard cell culture equipment and consumables cell culture medium, Fetal Bovine Serum (FBS), Trypsin mycoplasma detection system to verify the elimination success, e.g. Venor GeM mycoplasma PCR detection kit, cat. no. W 11-1025/1050/1100/1250) 3 Sample material The product is used for the elimination of all kinds of Mollicutes and related organisms (e.g. Mycoplasma, Acholeplasma, Spiroplasma, and Entomoplasma) in all kinds of cell and virus cultures. For best results the following recommendations should be considered: 1. Not more than 10 5 cells shall be used for a treatment to keep the mycoplasma load low. 2. The mycoplasmacidal activity of Mynox Gold is affected by the concentration of lipids and proteins in the reaction mixture, e.g. components in fetal calf serum supplement. These ingredients competitively bind Mynox Gold and prevent its binding to the mycoplasma membrane. A protocol was designed that requires a supplement of 5 % (v/v) fetal calf serum. Due to the mitigating effect of serum, it is impossible to design a specific protocol that is applicable or the treatment of biologicals with high protein and lipid concentrations. 3. The type of cell culture medium does not effect the efficiency of the treatment. Antibiotics, especially if required for selection, can stay in the elimination mixture. In rare cases cytotocicity might be increased by unpredictable interactions of the reagents. 4. Viruses shall be treated in combination with their host cells. 5. Mynox Gold does not penetrate the cellular membrane. Therefore, it cannot eliminate intracellular contamination. However, mycoplasma is an extracellular contaminate. Mycoplasma penetrans is the only species described intracellulary. Thus far, M. penetrans has not been reported as cell contaminant. 6. Since Mynox Gold works by biophysical means through association with the mycoplasma membrane, the reagent needs direct contact with the mycoplasma particle in order to be effective. Treatment of cell clusters should be avoided. Mycoplasma are protected in intercellular spaces as well as in pockets and clefts of the cell membrane, which can prevent contact with the drug. 4 Protocol guide These protocols have been designed for typical cell lines requiring standard media. The protocol can be used for adherent and suspension cell lines. It can not be guaranteed that these protocols will work in all laboratory situations. Modification of these protocols may be required per individual case. 1. mix in a sterile culture flasks or petri dish 4.5 ml standard cell culture medium with 5 % (v/v) FBS and 500 µl Mynox Gold starter treatment (vial with orange cap) 2. transfer 5 ml of 1x10 4 to 1x10 5 single cells in cell culture medium with 5 % (v/v) FBS into the mix. The total volume of the treatment mixture is 10 ml. Ensure the treatment of single cells (check under microscope!). If necessary, increase duration of Trypsin treatment or detach the cells from each other by pipetting up and down. Ensure that Mynox Gold is already present in the culture medium before adding cells. Add cells directly to the elimination mix to avoid evaporation (insert the pipette tip directly into the mix!) The final FBS concentration is 5 % (v/v). 3. grow the cell to 80-90 % confluency. 4. split the cells and passage at the usual rate. Add 500 µl of the Mynox Gold main treatment reagent (vial with transparent cap) to 9.5 ml of passaged cells in fresh media. Adjust the FBS concentration. Supplementation to 5 % is not required any longer. 5. repeat step 3-4 twice. After the third treatment (and a total of 4 passages starting with the starter treatment) the procedure is finished and the culture is free of mycoplasma. Fig. 1: Scheme of the treatment of adherent cell lines (applicable for suspension cells and adherent growing cells) 5 Testing for mycoplasma Mynox Gold treated cell cultures and virus stocks should be subcultivated for four additional passages without mycoplasma-active antibiotics and then assayed for mycoplasma reemergence to validate culture purity. For highly sensitive detection of mycoplasma contamination, we recommend Venor GeM mycoplasma detection kit that uses PCR technology (cat. no. 11-1025/1050/1100/1250). PCR detection methods should not be used instantly after treatment. Mynox Gold is lysing the mycoplasma particles and the mycoplasma DNA is released into the culture medium. This DNA would be detected by PCR giving false-positive results. Medium replacement and extracellular DNAses will reduce the level of free mycoplasma DNA within 1 to 2 passages.
