Health Effects Human Health Safety Assessment Effect Assessment Acute Toxicity Oral/inhalation/dermal Irritation / corrosion Skin/eye/respiratory tract Sensitisation Toxicity after repeated exposure Oral/inhalation/dermal Genotoxicity / Mutagenicity Carcinogenicity Toxicity for reproduction Not classified for acute toxicity for all three routes of exposure. Not irritating to skin and eyes. Not classified as sensitising to the skin, based on animal data. Low toxicity after repeated exposure by inhalation and oral route, not resulting in classification. At high concentrations, possible effects on the olfactory epithelium. No data on dermal repeated toxicity, but expected to be low owing to negligible skin absorption potential. Not classified either for mutagenicity or genotoxicity. No data available. No adverse effect on fertility and on development based on in vivo experimental study results. All these results are based on available data. Regarding toxicological hazard, dimethyl 2-methylglutarate is not classified according to EC 1272/2008 regulation criteria. Environmental Effects Environment Safety Assessment Effect Assessment Aquatic Toxicity Fate and behaviour Biodegradation Bioaccumulation potential PBT / vpvb conclusion Harmful to fish and algae. Not harmful to invertebrates. Readily biodegradable. Not potentially bioaccumulative. The substance is considered to be neither persistent and bioaccumulative and toxic (PBT) nor very persistent and very bioaccumulative (vpvb). Based on available data, dimethyl 2-methylglutarate is considered as harmful to aquatic life, but since it is readily biodegradable and not potentially bioaccumulative, it is not classified as dangerous for the environment, according to EU regulation (EC) 1272/2008. Exposure Human health Dimethyl 2-methylglutarate is manufactured in industry in closed processes which ensure that the risk is controlled. Where there is a risk of exposure to the substance, during (un)loading, sampling, analysis or maintenance operations and particularly in case of batch processes and spray applications, the exposure should be kept as minimum as possible by the use of appropriate risk management measures as suitable collective and personal protective equipment, good industrial hygiene practices and risk communication through appropriate training of workers. In end-use products like houshold cleaners or personal care products, dimethyl 2-methylglutarate is formulated to comply with requirements of the most stringent regulations for all applicable consumer uses to ensure that final products are used safely in the conditions of use written on the packaging. Environment Based on its physico-chemical properties, dimethyl 2-methylglutarate has a low potential for bioaccumulation and is readily biodegradable. It would be expected to partition predominantly into the aquatic compartment and not to adsorb on soil. Emissions in the air are not expected as the substance has a low potential for volatility. On industrial manufacturing and professional sites, effluents that may contain the substance must be directed to a waste water treatment plant. Due to its ecological properties, an indirect exposure of humans to dimethyl 2-methylglutarate via the environment is not expected. Risk Management Recommendations Human health For industrial and professional uses of dimethyl 2-methylglutarate and as recommended for the use of any chemical product, workers must be well informed and trained and must refer to the Safety Data Sheet (SDS). Where there is a risk of exposure to dimethyl 2-methylglutarate (during (un)loading, mixing, sampling, analysis or maintenance operations and particularly in case of batch processes and spray applications), it must be controlled by handling the substance under an adequate and efficient ventilation. Appropriate Personal Protective Equipment (PPE) must be worn (safety goggles, gloves, protective suit) as recommended in the SDS. In case of exposure to vapours or aerosol, a respirator with approved filter should be used. General industrial hygiene measures are required to ensure safe handling of the substance: Emergency equipment immediately accessible; use well-maintained PPE; wash hands and skin following contact; do not eat, drink or smoke at the workplace. For consumer uses, dimethyl 2-methylglutarate is formulated in suitable concentrations according to appropriate regulations, to ensure safe-use of the final products, in the conditions of use written on the product packaging.
