Challenge Nowadays, we are facing a growing and worrying phenomenon; more and more cities are concerned by pollution peaks due to the gradual increase of people living in urban areas and to the development of manufacturing companies. The skin is steadily exposed, additionally to sun rays, to various pollutants and particulate matter (PM) that increase cutaneous inflammation and contribute to accelerate cutaneous aging. In light of this alarming observation, ID bio has decided to carry on a new study on its latest active ingredient, Cell intact (INCI : Aqua (and) Polygonum Fagopyrum Seed Extract) already revealed for its antioxidant action, against UV and phytochemical aggressions. This ex vivo evaluation has been realized to prove the global efficiency against pollution to: -reduce the release of interleukin-8 (IL-8) that overreacts to create cutaneous discomfort -rebuild and reinforce the architectural epidermis network via the protection of tight junctions proteins (Claudins-1 and -4) to avoid premature aging signs A targeted pollutant in the atmosphere Pollution is global but from one city to another the cocktail of pollutants can be distinct in ambient air. In order to offer precise results, based on a pharmacological model, ID bio has decided to focus on a unique worldwide pollutant: benzo[a]pyrene or BaP. It is the major tracer of urban pollution due to PAHs (Polycyclic Aromatic Hydrocarbons), highly carcinogenic and mutagenic PM. This molecule that belongs to the VOCs (Volatile Organic Compounds) is quantified during pollution peaks as it is highly representative of the air quality.

BaP is mainly present in photochemical smog, the industrial fog soaring above polluted cities. It can be found in car exhaust gases and fumes, industrial smokes, tobacco, emanation of residential wood heating, etc. The botanical solution offered Protection of reactive skins IL-8 is an inflammatory cytokine that is released in case of harmful microbiological or chemical attacks inflicted to the skin. This autocrine mediator is particularly responsive to pollutants and that is over-released if skin is attacked. Cutaneous explants, and more precisely Normal Human Keratinocytes (NHK), have been stabilized and then topically pre-treated with 3 formulas (placebo, active at 1% and active at 3%) applied 24 hours before pollution with BaP. Then, another treatment with the 3 formulas has been carried out during 24 hours and the IL-8 dosage has been realized on supernatants (treatment media). The botanical active ingredient, Cell intact, enables to reduce the IL-8 release by -11,2% and 26,2 % compared to the placebo. Cutaneous hyperreactivity and diffuse redness are reduced in case of aggressions in urban area. Discomfort feeling (tautness, tingling, heating up, etc) is soothes. Thanks to the beneficial and significant action on IL-8, skin is less sensible and sensitive to pollution. Preservation of skin youthful appearance Tight junctions are established between cells and are composed of several families of transmembrane proteins, as claudins, occluding and adhesions molecules (ZO-1, ZO-2 ). A focus has been realized on Claudins-1 and -4 (CLDN1 and 4), located in the upper layers of epidermis (where keratinocytes differentiation takes place). Those key proteins actively participate in skin barrier protection by preventing small molecules entry and by keeping the epidermis integrity. In case of pollutants aggressions, homeostasis is disrupted and this barrier is damaged. Calcium flux is increased and skin architectural network is tumbled out which increases risks of premature aging signs arrival. An immunostaining on NHK has been carried out on skin explants, stabilized and then topically pre-treated with 3 formulas (placebo, active at 1% and active at 3%) applied 24 hours before pollution with BaP. Then, another treatment with the 3 formulas has been carried out during 24 hours and the expression of CLDN1 and 4 has been observed with fluorescent immunostaining. This technique permits to observe the expression profile of the proteins in the epidermis. We noticed that, without Cell intact, network of tight junctions is modified, less structured and dense (fluorescence -) whereas in presence of the active ingredient, the architectural network is re-established and tightened compared to polluted control (fluorescence +). The product enables to increase cells cohesion of superficial epidermal layers and to reinforce tissue barrier function. Skin is more resistant to environmental attacks and premature aging signs are kept away.

Those new results show a larger scope of intervention against pollution and the creation of a shield against aggressions.
