5 Context The laboratory of Virology is a National Reference Center for arboviruses and influenzavirus Existence of a virological and serological surveillance of Dengue virus and other arboviruses in French Guiana Flavivirus: Yellow fever, Saint-Louis encephalitis, West Nile virus Alphavirus: Tonate (VEE complex), Mayaro, Chikungunya Bunyaviruses from South America Existence of a virological surveillance of influenzavirus circulating in the French Departments of the Americas (Martinique, Guadeloupe and French Guiana) 5

6 Missions To provide a virological expertise on circulating dengue strains in French Guiana and French West Indies (Martinique, Guadeloupe) To participate to development and/or validation of new diagnostic tools To contribute to the epidemiological and virological surveillance of Dengue in relation with Public Health authorities from French Guiana Improvement of existing surveillance system Reinforcement of alert Identification of the circulating dengue serotype Evolution of outbreak 6

7 Diagnostic tools Virus isolation Cell culture (mosquito cells AP 61, C6/36) and IF for identification of serotype Genome detection RT-PCR (Lanciotti primers) Real-time PCR in progress Antigen detection ELISA capture assay directed to the NS1 antigen Serological diagnosis Specific capture of IgM (MAC-ELISA test) and IgG (ELISA) Limitations of serological tests In areas where other flaviviruses are circulating and in individuals previously vaccinated with YF (or JE) vaccine 7
