2 New products VitaFast Vitamin C (L-Ascorbinsäure / L-Ascorbic Acid) Ascorbic acid, also known as Vitamin C, is used as a vitamin but also as an antioxidant and preservative in the food industry. A condition called Scurvy can manifest itself in people with a lack of vitamin C. As vitamin C is partially converted into the equally biologically active dehydroascorbic acid in food, both substances should be detected in one test system. The VitaFast Vitamin C (L-Ascorbinsäure/ L-Ascorbic Acid) test (Art. No. P1010) is an enzymatic test in microtiter plate format for the quantitative determination of vitamin C (L-Ascorbic acid) in foods, pharmaceutical products and other sample materials. This test also allows the important determination of the total content of vitamin C (L-ascorbic acid + L-dehydroascorbic acid). The test kit contains an L-ascorbic acid standard for calibration and all necessary reagents sufficient for 50 determinations (including 4 standard series) to be carried out with the test. A microplate photometer ( nm) is necessary for the analysis. The limit of detection and quantification of the VitaFast Vitamin C test is 7.8 mg/100g (ml) with a recovery rate of between % (if spiked with freshly prepared ascorbic acid). The test has been validated extensively. The table shows some results of various spikes in different food matrices. The samples were spiked in duplicate or triplicate and extracted according to the product information. VitaFast Vitamin C (L-Ascorbinsäure / L-Ascorbic Acid) Art. In order to meet the market and customer demands, this new vitamin C test has been introduced as a supplement to the VitaFast line. The test is suitable for customers who have access to a microplate photometer (wavelength: nm) and do not carry out the classic, colorimetric determination in 1 cm cuvettes with the spectral photometer. 3 RIDA COUNT Total 24 R-Biopharm has been offering a new, innovative product from the RIDA COUNT line since the middle of September The RIDA COUNT Total 24 (Art. No. R1011) is a test card which allows the user to determine the total aerobic bacteria count of food samples or contact tests within 24 hours. The test cards are developed in such a way that they can be always incubated at 35 C independently of the nature of the sample. The detection after only 24 hours is made possible by the special chromogen included in the test card. As soon as the microorganisms start to grow after application of the sample onto the test card, the chromogenic substance is dissociated by microbial enzymes of glycolysis and of the citrate cycle and a dark red coloring builds up. The colony developing is therefore made visible in the shortest amount of time. The new product information and further information on the new RIDA COUNT Total 24 is available via R-Biopharm. Our products RIDA COUNT use for cosmetics Following increased customer demands about whether respectively how cosmetics can be tested with RIDA COUNT test cards, R-Biopharm has developed a sample preparation protocol for cosmetics of different consistencies. The protocol for the preparation of cosmetics for application on RIDA COUNT test cards is relatively easy to handle and only deviates slightly from the process instructions for the microbiological analysis of cosmetics (Guidelines for microbiological Quality Control of Cosmetics). Compact Dry X-BC validated and certified The microbiological test Compact Dry X-BC (Art. No. HS9721, 100 tests and HS9722, 40 tests) for the determination of the number of Bacillus cereus in food samples has been certified as a validated method since end of August. The corresponding validation studies were carried out by the MicroVal validation authority according to the ISO Guidelines. The Compact Dry X-BC method was compared with the ISO Standard 7932:2004 for the quantitative determination of Bacillus cereus. MicroVal s validation certificate confirms that the comparison of the ISO method with the alternative method (Compact Dry X-BC) shows that no significant difference could be determined. Therefore, Compact Dry X-BC was recognized as a suitable method for the quantitative determination of B. cereus in food. 4 News from our cooperation partner GEN-IAL GmbH Juice analysis with real-time PCR first Alicyclobacillus PCR Screening Kit Alicyclobacillus is a strictly aerobic and spore forming spoilage bacteria. It is especially important for the fruit juice industry, where it has been shown to produce high amounts of guaiacol imparting off-flavors in fruit juices. Early screening of Alicyclobacillus is very important for the fruit juice canning industry because common pasteurization techniques do not deactivate the spores. A contamination can lead to considerable loss of quality and subsequently to economic losses for the producers. Alicyclobacillus acidoterrestris is considered the most important spoilage species within the Alicyclobacillus genera, but others e.g. Alicyclobacillus acidocaldarius have also been isolated from spoiled products. General culture methods take days to detect the presence of Alicyclobacillus species in samples, a further 24h to confirm guaiacol production and identification of isolates require even longer. To overcome this difficulty, GEN-IAL developed a multiplex real-time PCR method to detect and differentiate Alicyclobacillus species in fruit juices and concentrates much earlier. First-Alicyclobacillus multiplex PCR kit (Art. Information from R-Biopharm Rhône, Scotland Method for Detection of Chloramphenicol in Red Wine by LC-MS/MS In recent weeks the European Rapid Alert System for Food & Feed (RASFF European Rapid Alert System for Food & Feed*) has identified residues of the antibiotic chloramphenicol in various imported enzyme products intended for use in the Food and Animal Feed Industry. It is believed that this incident was caused by the outsourcing of a chloramphenicol contaminated pectinase enzyme preparation which was then imported into the EU directly. Since chloramphenicol is a banned substance, industry trade bodies within the EU, including the Association of Manufacturers and Formulators of Enzyme Products (AMFEP Association of Manufacturers and Formulators of Enzyme Products) have subsequently taken positive action to test contaminated products and attempt to remove the possibility of them entering the food chain and endangering human health. Although the enzymes are used in very low amounts, traces of chloramphenicol have already been found in red wine in countries including Spain. The Spanish Reference Lab for wine analysis has thanked R-Biopharm for its rapid response to industry demands to test for chloramphenicol residues in wine where contaminated enzymes could have been used during the production process. A new, simple and fast sample clean-up procedure by immunoaffinity column has been proposed to allow confirmation of this residue by LC-MS/MS analysis at the EU MRPL (Minimum Required Performance Limits) of 0.3 µg/l (ppb) using EASI-EXTRACT CHLORAMPHENICOL (Art. No. RBRP300/ P300B): take 10 ml wine and dilute with 10 ml water check ph and buffer to 7.4 if necessary with sodium hydroxide filter the diluted sample through a Whatman No. 113 or No. 4 filter paper or centrifuge at 4000 rpm for 10 min continue on page 5 * page 4
5 continued from page 4 apply all 20 ml diluted wine to the EASI-EXTRACT CHLORAMPHENICOL immunoaffinity column at 2 ml per min or under gravity (a slow steady flow is necessary for the antibody to capture all of the antibiotic present in the sample) wash the immunoaffinity column with 10 ml water at a rate of 5 ml per minute pass air through the immunoaffinity column using a syringe to gently remove any residual liquid elute drop wise with 2 ml 100 % methanol and collect in a glass tube (back-flushing of the eluate is recommended) inject 10 µl onto the LC-MS/MS system Initial collaborative customer results for Spanish red wine are promising however please note that these results were based on a draft protocol and a full validated method for wine will follow. 0.3 µg/l - 72 % recovery of CAP 0.5 µg/l - 73 % recovery of CAP 2.0 µg/l - 93 % recovery of CAP Other validated application notes are available on request for chloramphenicol in milk, eggs, meat, honey and seafood.
