You and your research staff could spend years learning to overcome these problems or you can try a Burleigh PCS-5000 Series Micromanipulator and learn what total control really means. Total control means your fingertip commands are instantly and precisely converted to smooth and predictable pipette motion without drift. Only the EXFO piezoelectric micromanipulators provide this capability, since our approach avoids some of the inherent difficulties of other technologies. Hydraulic manipulators experience drift due to fluid expansion, contraction and leakage. Mechanical manipulators transmit fingertip vibrations to the pipette and do not always move in predictable directions. Motorized lead screws can produce a jerky motion at slow speeds and, when direction is reversed, backlash delays motion and makes operation unpredictable. EXFO s solid-state piezoelectric (PZT) design produces extremely smooth and predictable motion and negligible drift. Your position commands are instantly and precisely converted to pipette movements with no backlash. The ceramic and metal kinematic design of the EXFO system offers the lowest possible drift. Leakage, backlash, compliance and drift of hydraulic, mechanical and motorized lead screws are a thing of the past. Only EXFO offers the solid-state stability of piezoelectric technology in complete and easy-to-use products for Life Science research. PZT actuators are solid ceramic structures that expand under applied voltage. When controlled by EXFO s drive electronics and ergonomic control interface, extremely precise, fast, stiff and stable positioning is easily achieved. The position of each PZT actuator is determined by the CD voltage applied by the power supply. EXFO s linear lownoise electronics and cables are fully shielded and carefully grounded to produce negligible interference of your recordings. Scientists around the world are successfully using EXFO s manipulators for the most sensitive single channel recordings. PZT actuators produce negligible internal thermal drift. When external thermal changes are considered, an EXFO system contributes the same or less thermal position drift than other components such as the microscope, mounting structure and headstage. With proper temperature control, pipette drift of less than one micrometer per hour can be achieved. No other micromanipulator has inherently lower drift than an EXFO micromanipulator. LINEAR STAGE WITH PZT FLEXURE ASSEMBLY The heart of the system is the PZT Flexure Assembly which is integrated into the linear stage. The stage is constructed from aluminum, utilizes cross roller bearings and provides manual coarse control through a 32 thread-per-inch lead screw with a range of 25mm. Fine control is provided through the PZT flexure with optional ranges of 150 or 300 micrometers and a minimum resolution of 60 nanometers. The stage can be provided without the PZT element if only coarse control is required.

3 PCS-520 Micromanipulator Assembly OPERATIONAL FEATURES THAT MAXIMIZE YOUR PRODUCTIVITY The PCS-520 Micromanipulator Assembly is comprised of three linear stages, two rotary stages with lockable stops, one set of angle brackets and a headstage mounting plate. Each Micromanipulator Assembly also includes a Model PCS-500-MB Magnetic Base for mounting to a ferromagnetic surface. Each linear stage of the PCS-520 Micromanipulator Assembly utilizes cross roller bearings that form a stiff and precise single axis bearing system with high load capacity and minimal friction. The solid-state nature of PZT actuators produces virtually zero heat while providing 60 nm resolution, zero backlash and negligible inherent drift. Headstages and pipettes are easily installed using the Headstage mounting plate. Axon CV-203B and HEKA EPC-9 Headstages can be bolted directly. Other headstages can be mounted to the flat plate by adding the appropriate mounting holes. The pipette angle of approach can be easily optimized using the headstage pivot. This feature makes setup easy on an inverted or upright microscope, especially when space or working distance is critical. Any angle can be selected and easily repeated using the integral adjustable stop mechanism. Extremely steep or shallow angles can be accomplished using the PCS-500-SSH Steep/Shallow Headstage Adapter. This adapter is recommended when using the PCS-520 Micromanipulator Assembly with upright microscopes with water immersion objectives. The symmetric and universal mechanical design of the Micromanipulator Assembly allows easy customization. The assembly can be modified to provide left sided or orthogonal approach axis operation with minimal effort. No special adapter kits are required.

4 PCS-5000 Series ROTARY PIVOT STAGES WITH ADJUSTABLE SET STOPS ALLOW EASY PIPETTE EXCHANGE Quick and easy pipette exchange is accomplished using the two rotary pivot stages with adjustable stop mechanisms. The adjustable stop mechanism, found in both the headstage rotary pivot stage and base rotary pivot stage, consists of a brass block, a locking ring, two set screws and a pin. By loosening the set screws, you can rotate the locking ring until the pin rests against the brass block. Tightening one of the set screws with the pin in this position gives a precise and repeatable home position. Once the set stops are adjusted to your working conditions, you can change pipettes easily and return to the field of view of the microscope as shown in the pipette exchange figure. To begin, loosen the pivot knob on both the headstage pivot stage and base pivot stage. Rotate the pipette up and out from the microscope objective. See figures 1 and 2. The headstage and pipette are now in the forward position and very accessible for pipette exchange. See figure 3. You can temporarily tighten the headstage pivot knob to hold the headstage steady while you exchange the pipette. After pipette exchange, rotate the pipette back under the objective as far as it will go in both the horizontal and vertical planes. The pre-set mechanical stops (described above) define the original home position. After tightening the pivot knob on both the headstage pivot stage and base pivot stage, and allowing for some variation in pipette lengths, you should be back in home position and ready to proceed. 5 PCS-5000 SERIES PATCH CLAMP MICROMANIPULATOR SYSTEM CONFIGURATIONS PCS-503 Axis Control Unit You can achieve total control of your pipette position using fingertip commands to the PCS-503 Axis Control Unit (ACU). Each of the three linear stages with PZT fine control has a cable (not shown) that connects to any one of the three ports on the Axis Control Unit. There is total flexibility in choosing which knob you would like to control which linear stage. Simply rotate the larger diameter knobs which are connected to precision three-turn potentiometers. Three full rotations of the potentiometers correspond to full range of the PZT flexure assembly. The position resolution is 0.04% of the total PZT travel, providing 60 nm of resolution with a 150 µm PZT stage. This means you can move the pipette in smaller increments than can be detected through your microscope! A key feature of the ACU is adjustable friction potentiometers. Using simple adjustment procedures, you can easily optimize the potentiometer friction to the resistance you desire. PCS-503 Axis Control Unit The PCS-PS60 Power Supply provides a regulated, very low noise, constant, 60 volt output for the Axis Control Unit. The power supply can be shared by two micromanipulator / ACU systems to minimize cost and space consumption when multiple manipulators are required on the same microscope. Input power is 100 volts to 240 volts and 50/60 Hz ensuring world wide compatibility. In addition, the PCS-5000 System is compliant with strict European Community (CE) electronic regulations. 6 PCS-5000 Series ACCESSORIES TO FURTHER ENHANCE YOUR PRODUCTIVITY PCS-500-SSH Steep / Shallow Headstage Adapter The PCS-500-SSH Steep/Shallow Headstage Adapter adds unprecedented flexibility for positioning the headstage. It allows the orientation of a headstage and pipette holder at very steep or shallow angles. And it further simplifies pipette exchange even when the pipette is in the tightest places. Its symmetrical design offers these capabilities from both the right and left side of the microscope. Steep pipette angles are often desired to minimize the length of the electrode in solution with the typical angle being greater than 45 degrees. The shorter the length of the pipette in the solution, the lower the stray capacitance and the lower the electrical noise measured while recording. On a typical inverted microscope, the maximum approach angle is limited by the interference of the headstage with the condenser. The Steep/Shallow Headstage Adapter holds the headstage so that clearance with the condenser is maximized. Shallow pipette angles are typically needed when working with brain slices on upright, fixed-stage microscopes. The optimum angle allows an electrode to be moved in a straight line over the brain slice chamber and under the microscope objective without breaking the pipette. The typical angle is 15 to 25 degrees. The Steep/Shallow Headstage Adapter extends the headstage closer to the brain slice chamber and away from the manipulator. This extended design also makes it easier to arrange several micromanipulators around one brain slice. When working with a pipette close to the condenser or objective of the microscope, swinging the micromanipulator up and out by hand can be a challenge. The PCS-500- SSH Steep/Shallow Headstage Adapter integrates a quick and efficient mechanism that allows you to retract the pipette up to three inches from the preparation and then easily return to home position after pipette exchange. PCS-500-EB Extension Bracket Kit Even though the PCS-5000 Micromanipulator is designed for maximum flexibility, there may be a time when you need to give it a little boost to achieve perfect pipette position. In this case, you can use the PCS-500-EB Extension Bracket Kit. The kit includes three spacer discs that are three inches in diameter and one-half inch thick. The extension plate is fastened to the mounting surface with one or all of the discs. PCS-500-MB Magnetic Base The PCS-500-MB Magnetic Base can be used to mount to a magnetic surface. This allows for quick positioning, attachment and detachment of the micromanipulator to a magnetic surface. PCS-500-EB Extension Bracket Kit
