16 Abstrakty SEKCE: PTAČÍ SCHISTOSOMY CERCARIAL DERMATITIS: AN UPDATE OF TRANSMISSION AND SPREADING OF BIRD SCHISTOSOMES P. HORÁK Department of Parasitology, Faculty of Science, Charles University in Prague, Viničná 7, Prague 2 Cercarial dermatitis is a hypersensitive skin reaction (disease) caused by schistosome larvae - cercariae; it is restricted to aquatic environment. Besides mammalian and human schistosomes (Schistosoma spp. and other genera), avian schistosomes are frequently reported as the causative agent. In some areas of the world cercarial dermatitis is regarded as an (re)emerging disease, because it appeared newly (no reports in the past) or the number of human cases (water reservoirs with schistosomes) increased; this applies to e.g. Chile, the United Kingdom, the United States and Denmark. Global climate changes may contribute to dissemination of schistosomes by birds and snails, accelerated development of parasites, increased contact of people with the causative agent, etc. Bird schistosomes require water snails for their larval development. The prevalence of infections in snails is usually low (below 1%), but in some locations/under certain circumstances it may reach up to 50%. These differences are, at least partly, influenced by the local spectrum of snail species and their susceptibility to infection. Unfortunately, proper identification of some snail species and our knowledge of the mechanisms determining snail susceptibility/resistance may represent limits of understanding transmission modes. Birds as definitive hosts are frequently parasitized by schistosomes, and anseriform birds seem to play a dominant role. In some localities, the prevalence of schistosomiasis in birds may reach up to % (e.g. New Zealand, the United States). The intensity of transmission to birds is poorly studied, but we know from experimental infections that at least two ways of infection exist: percutaneous (penetration of the bird skin) and peroral (penetration of the bird mucosa). Although there is limited data about natural infections, bird schistosomes seem to be pathogenic in some cases, and their development/growth probably depends on host physiological suitability. 16

Cathepsin B1 je lokalizován ve střevě schistosomul a dospělců a je velmi pravděpodobné, že se účastní trávení složek nervové tkáně a krve hostitele. Cathepsin B2 byl nalezen v postacetabulárních žlázách cerkárií a zřejmě napomáhá při penetraci do hostitele. V naší práci jsme se zabývali biochemickou charakterizací rekombinantních forem těchto peptidáz. Rekombinantní pro-cathepsiny B (izoformy TrCB1.1 a TrCB1.4 a dále TrCB2) byly získány expresí v systému metylotropních kvasinek Pichia pastoris. TrCB2 byl schopen autoaktivace za podmínek expresního media, zymogeny pro-trcb1.1 a pro-trc1.4 byly efektivně aktivovány experimentálně po inkubaci s ionexovou matricí Macro-Prep-High- S Support (Bio-Rad) či s pepsinem. U aktivovaných rekombinantních cathepsinů B byla potvrzena exopeptidázová aktivita pomocí specifického syntetického substrátu Hippuryl-His- Leu-OH. Dále byla určena ph optima jejich endo- i exopeptidázových aktivit a ověřena schopnost štěpit vybrané přirozené proteinové substráty (složky nervové tkáně, krve a kůže) - myelin basic protein, fibrinogen, albumin, IgG, kolagen a částečně i hemoglobin. Následně byla pomocí MALDI-MS/MS, LC-MS/MS identifikována štěpná místa v substrátech myelin basic proteinu a hemoglobinu vzniklá po působení těchto enzymů. Byly nalezeny drobné rozdíly ve vlastnostech studovaných enzymů, které budou diskutovány v kontextu jejich biologických aktivit. Poděkování: Czech Science Foundation grant no. 206/09/H026 17

18 Abstrakty ODVRHOVÁNÍ GLYKOKALYXU U CERKÁRIÍ PTAČÍCH SCHISTOSOM TRICHOBILHARZIA SPP. L. MIKEŠ & J. CHALOUPECKÁ Katedra parazitologie, Přírodovědecká fakulta, Univerzita Karlova v Praze, Viničná 7, Praha 2 Během penetrace do definitivního hostitele se cerkárie schistosom transformují na stádia zvaná schistosomuly. Tento proces je doprovázen odhozením ocásku a vyprazdňováním penetračních žláz. Tělo cerkárie přitom prochází četnými ultrastrukturálními a molekulárními změnami, které se týkají i povrchu tegumentu. Jako příklad lze uvést tvorbu dvojité membrány a ztrátu silné vrstvy vysoce imunogenního glykokalyxu, který pro cerkárie představuje ochrannou bariéru ve vodním prostředí. Glykokalyx schistosom má specifické složení sacharidových molekul, které jsou vázány k lipidům nebo proteinům membrány tegumentu. O mechanismu jeho odvrhování je poměrně málo informací, hypotetická je možnost indukce tohoto procesu proteolytickými enzymy z penetračních žláz. Důležitou součástí našeho projektu je charakterizace změn v glykosylaci povrchu cerkárií při transformaci na schistosomuly pomocí vazby fluorescenčně značených lektinových prób a protilátek proti vybraným sacharidovým epitopům. Hlavním cílem experimentů je odhalit mechanismus a ukázat průběh odvrhování glykokalyxu u cerkárií ptačích schistosom rodu Trichobilharzia a objasnit případný podíl produktů penetračních žláz na tomto procesu. K tomuto účelu jsou mj. využívány rekombinantní formy žlázových enzymů cerkárií. Zároveň probíhá analýza proteomu penetračních žláz Trichobilharzia regenti a testy zapojení exogenních molekul původem z hostitelů na indukci odvrhování glykokalyxu. 18

19 X. české a slovenské parazitologické dny 2012 HOST-PARASITE INTERACTIONS OF BIRD SCHISTOSOMES (TRICHOBILHARZIA SP.) AND IMMUNOMODULATED MICE M. CHANOVÁ Institute of Immunology and Microbiology, 1 st Faculty of Medicine, Charles University in Prague, Studničkova 7, Prague 2, Czech Republic Bird schistosomes of the genus Trichobilharzia are able to penetrate the skin of mammalian host and, in previously uninfected mammals, to migrate, feed on host tissues and partially develop. The routes of migrating larvae (schistosomula) and the pathogenic impact on invaded tissues in abnormal mammalian host (mice) are known for species of T. szidati, visceral species migrating via blood system, and T. regenti, neurotropic nasal species migrating through peripheral and central nervous tissue. The present study extends the knowledge on pathogenic impact on mammalian host with modulated immune system. For experimental infections, BALB/c mice were immunomodulated by complete and incomplete Freund's adjuvant 7 days prior to exposition to cercariae (subcutaneous injection of 100 µl of adjuvant; subsequent infection by immersion of foots into the water containing cercariae). Control mice remained uninfected (BALB/c), or were infected without previous immunomodulation (BALB/c; SCID). Mice were euthanized after desired period (1-15 days) post infection (p.i.). The spinal cord and brain was fixed with 4% paraformaldehyde and embedded in paraffin or JB4 embedding medium. Other organs/tissues were torn and searched for parasite presence under binocular stereomicroscope. The migratory route/parasite location and local immune cell reaction were studied by methods of histology and immunohistochemistry, systemic immune response (T h 1 vs. T h 2 oriented reaction) was characterized by flow cytometry. 20 Abstrakty TRICHOBILHARZIA REGENTI: PATHOGENIC EFFECT OF SCHISTOSOMULA MIGRATION L. LICHTENBERGOVÁ & P. HORÁK Department of Parasitology, Faculty of Science, Charles University in Prague, Viničná 7, Prague, Czech Republic Trichobilharzia regenti, similarly to other bird schistosomes, represents causative agent of skin inflammatory reaction, cercarial dermatitis. Moreover, T. regenti is known as a parasite with neurotropic mode of behavior. Due to migration through the host nervous tissue, T. regenti infections of specific bird and accidental mammalian hosts could represent a more serious health risk than those by other species of bird schistosomes. The infection of vertebrate host starts in water environment when cercariae (infective stage) penetrate the host skin. Successfully penetrating cercariae undergo cercaria/schistosomulum transformation with reconstruction of the tegumental surface. Shedding of antigen rich glycocalyx increases the resistance to host immune system. Despite surface changes, T. regenti infection of the skin of bird and mammalian hosts leads to the development of inflammatory reaction. Study on primo-infected ducks showed that the tissue in vicinity of schistosomula was infiltrated by heterophils, mast cells, CD3 lymphocytes and macrophages. Nevertheless, the immune reaction was only mild and schistosomula were able to escape from the skin and migrate through/along the peripheral nerves to the CNS. Histological observation revealed that the presence of schistosomula in the duck CNS initiated an influx of immune cells (heterophils, CD3 lymphocytes, macrophages) to the exposure sites, and proliferation and activation of astrocytes. Schistosomulum migration caused a disruption of axons and subsequent degeneration of neurons. Parasites were frequently observed in the subarachnoidal space of the spinal cord and, contrary to the persisting inflammation, schistosomula survived longer in this area than in gray or white matter of the spinal cord. Particular factors/antigens responsible for glial cells activation and parasite destruction are unknown, but their characterization is in focus. In the case of schistosomes, freely swimming miracidia recognize different stimuli such as MAGs substances (miracidia attracting glycoproteins) released by snails before they are attacked by schistosomes. Then, snail immunity is one of important factors determining compatibility or incompatibility of snails and trematodes. An incompatible parasite is usually eliminated by immune reactions in which hemocytes represent the main effector cells. On the other hand, a compatible trematode probably modulates activity of immune cells in order to avoid attack. In our experiments, the snails of Radix lagotis were infected experimentally by miracidia of the bird schistosome Trichobilharzia regenti. Histological evaluation of infected individuals showed that hemocytes were attracted by schistosome larvae at intervals 2, 3, 5, 12, 16 and 20 hours post infection (p.i.). Then, the number of hemocytes gradually decreased from 20 to 44 hours p.i., and later (60 and 92 hours p.i.) no hemocytes were observed around the larvae, suggesting that cellular reactions might be supressed by parasites. Some activities of hemocytes derived from infected (patent period) and noninfected snails were compared. Phagocytic activity of hemocytes from infected snails dropped to about 50% if compared to hemocytes from noninfected snails, whereas nitric oxide production increased by 130% in infected snails vs. 100% in nonifected snails after 60 minutes of measurement. It seems that also activities of selected enzymes comprising hemocyte signalling pathways may be affected by schistosome larvae. The data can contribute to a better understanding of trematode-snail compatibility.
