1 Protocol (PRO) Cell Culture Protocol for Hydrogel 2D and 3D Cell Culture PRO/BGX/001 Materials Provided Gel solution (stiffness of Gel will vary as per type of product requested), packaged in a glass vial. Materials Not Provided Cell culture multi-well plates and inserts. Tissue culture media. 2 1. GEL PREPARATION METHOD Prior to use, place the cell media and Peptide Gel solution(s) in an incubator or water bath for 30 minutes until the solution(s) reaches 37 C. NOTE: It is advisable to remove the required volume of gel solution into a sterile container or plating up in well plate while incubating at 37 C. GENTLY mix the Peptide Gel solution(s) using a pipette to produce a homogeneous solution. During mixing the pipette tip should not be removed from the solution. If air bubbles are present in the solution, remove these by placing the solution in a bath sonicator for 30 seconds. 2. 2D CELL CULTURE METHOD 2.1 2D Cell Culture Method (Well Format) Pipette the Peptide Gel solution into the bottom of the well plate. Volumes of Peptide required are shown in Table 1 for each plate size used. Refer to diagram below for guidance. 1.5 hours at 37 C and 5 % CO 2 Place the well plate containing the Peptide Gel solution in the cell incubator at 37 C with a humidified atmosphere of 5 % CO 2 for 1½ hours to form the Gel. After 1½ hours of incubation, GENTLY pipette cell media onto the surface of the Gel (Refer to Table 1 for cell culture media volumes). Then, place the culture plate in the incubator overnight without adding cells. Minimum time of incubation before cells can be added is 6 hours. 3 Please note that the cell number stated is a recommended guideline only and can be adjusted depending on the cell type or application. After overnight incubation, gently remove the cell media from top of the Gel and replace with the prepared cell suspension. Cell suspension volumes required are shown in Table 2 for each plate size used. Table 2: Volumes of Cell Suspension required for different well plate sizes using 2D cell culture in well format. Volume of Prepared Cell Suspension (µl) on top of Gel 96 Well Well Well D Cell Culture Method (Insert Format) Pipette the required volume of Peptide Gel solution into each well plate insert. Volumes of Biogelx Peptide Gel solution and cell culture media required are shown in Table 3 for each plate size. Add cell media into each well outside of the insert, so that the Peptide Gel solution in each insert is in contact (throu gh the membrane) with media from underneath. Refer to diagram below for guidance. Insert Place the culture plate containing the inserts, media and Peptide Gel solution in the cell incubator at 37 C with a humidified atmosphere of 5 % CO 2 for 1½ hours to form the Gel. After 1½ hours of incubation, GENTLY pipette cell media onto the surface of the Gel (Refer to Table 3 for cell culture media volumes) and replace the surrounding media outside the insert with fresh cell media. Then, place the culture plate in the incubator overnight without adding cells (minimum time of incubation before cells can be added is 6 hours). PRO PRO/BGX/ Page 3 of 7 06 Jan 2015

Remove the old media from the Gel surface within the insert making sure the pipette tip does not touch the surface of the gel whilst doing so. GENTLY pipette the prepared cell suspension onto the top of the gel in each insert. Replace the old cell media outside the insert with fresh media. Volume of prepared Cell Suspension (µl) on top of Gel Media (µl) added outside the insert 24 Well Well Well D CELL CULTURE METHOD 3.1 PEPTIDE GEL AND CELL PREPARATION Trypsinise cells and centrifuge so as to obtain a cell pellet. Determine the cell density. Remove the required volume of cells to a sterile tube to give a final cell density of x 10 6 cells/ml in Peptide Gel solution. Centrifuge the cell-media mix for 5 minutes at 1500 rpm to again obtain a cell pellet and remove media, leaving a maximum of 10 % media in which to re-suspend the cell pellet. Disturb the pellet through heavy shaking to allow breaking up of the cell mass. To this, add the required volume of Peptide Gel solution (0.5 1 x 10 6 cells/ml of Peptide Gel solution). 5 Mix by gently pipetting up and down to allow even distribution of cells in the Peptide Gel solution. Ensure the pipette tip does not leave the Peptide Gel solution when mixing as this can introduce air bubbles into the gel structure. After 1 hour of incubation, add media GENTLY to the surface of the Gel ( Table 6). Replace the media on the surface of the gel after 2 hours with fresh media. 6 Incubate the prepared well plates at 37 C with a humidified atmosphere of 5 % CO 2. After 24 hours of incubation, replace the media with fresh media. Care should be taken when removing the media on top of the gel; to avoid direct contact with the gel, carefully place the pipette tip at the top of the media and remove the media slowly D Cell Culture Method (Insert Format) Add the prepared Peptide Gel/cell solution to each well plate insert required (ensure gentle pipetting). Add cell culture media into the well (outside the insert) and incubate at 37 C with a humidified atmosphere of 5 % CO 2 for 1 hour. Volumes for Peptide Gel/cell solution and media for each culture plate size are shown in Table 7. Refer to diagram below for guidance. Volume of Peptide Gel/Cell Solution per Insert (µl) Media (µl) outside the insert 24 Well Well Well After 1 hour of incubation, add media GENTLY to the surface of the Gel (Table 8) and replace the surrounding media with fresh media (Same volumes as shown in Table 7). Replace the media on the surface of the gel after 2 hours with fresh media (Table 8). Media added to the gel surface after 1 and 2 hours of incubation (µl) 24 Well Well Well 1000 Incubate the prepared well plates at 37 C with a humidified atmosphere of 5 % CO 2. After 24 hours of incubation, replace media with fresh media (surface of the Gel and outside the inserts). Care should be taken when removing the media on top of the gel; to avoid direct contact with the gel, carefully place the pipette tip at the top of the media and remove the media slowly. 7 D Cell Culture Method (Sphere Format) Add cell media to each of the desired wells in the required well plate ( Table 9 shows volume required for each well plate size). Then GENTLY add the Peptide Gel/cell solution to each well, using a pipette in which the tip has had 1 cm of the end cut from it. This allows for the Peptide Gel/cell solution to be delivered to the well in a ball-like structure. Pipette tip should be fully immersed before Peptide Gel/cell solution is released into the media, do not add drop-wise. Refer to diagram below for guidance. Incubate the prepared well plates at 37 C with a humidified atmosphere of 5 % CO 2 for 3 hours.
