3 INTRODUCTION 1. BACKGROUND This manifold is suitable for vacuum filtration with standard 96-well filter plates. It can work with assays using the Firefly particles but is also compatible with non-firefly assays. Packaged complete with vacuum control assembly: Gauge indicates vacuum level applied to filter plate. Teflon flow restrictor and relief valve limit vacuum to assaycompatible levels. If higher vacuum levels are desired, these components are removable. Fine vacuum control enabled via set-point knob. Included 3/8 ID tubing to interface with end user s vacuum trap. 2

4 GENERAL INFORMATION 2. PRECAUTIONS Please read these instructions carefully prior to Assembling, Calibrating or Using the equipment. All components have been formulated and quality control tested to function successfully. Modifications to the components or procedures may result in loss of performance. 3. STORAGE Upon receipt of this equipment, please store at room temperature. Refer to list of materials supplied for storage conditions of individual components. 4. 5 GENERAL INFORMATION 6. LIMITATIONS Modifications to the components or procedures may result in loss of performance. 7. TECHNICAL HINTS For best results with Firefly particles maintain vacuum around -3 in Hg. Never allow vacuum stronger than -5 inhg. When filtering Firefly hydrogel particles, take care that particles are not dry for more than a few seconds. If plate does not filter, press firmly on the plate and check that the gauge reads non-zero. To use a partial plate, tape or seal off any non-assay wells to maintain even vacuum across the plate. 4

6 ASSAY PREPARATION 8. VACUUM MANIFOLD ASSEMBLY Keep all components at RT during preparation. 8.1 Connect waste output on vacuum control assembly to a vacuum trap using your own fitting (3/8 ID tubing included). 8.2 Insert supplied manifold tube into push-to-connect fitting on vacuum control assembly (use enough force such that tubing does not fall out). Manifold tube is released from assembly by holding the colored ring against the push-toconnect fitting and pulling on the tubing. 5

7 ASSAY PREPARATION 9. VACUUM MANIFOLD CALIBRATION Vacuum Manifold PLUS contains three pieces a vacuum control assembly, a lower base, and a top frame. Place the top frame onto the base, taking care to ensure the top frame is evenly seated on the manifold base. Place a filter plate lid on top of the manifold frame and turn on the vacuum source. Gently press down on the sides of the lid to ensure a vacuum seal. Adjust the set-point knob on the vacuum control assembly until the gauge needle is around -5 in Hg. Turn off the vacuum source and note the time it takes for the lid to be released from vacuum. If this time is more than 3-5 seconds, slightly open the setpoint knob and repeat steps 1-3 until this is achieved. Readings around -3 inhg are also acceptable. 6

8 ASSAY PROCEDURE 10. VACUUM MANIFOLD USE 10.1 This step is only necessary for filtrate collection. Skip to Step 2 if your filtrate is waste. Remove the vacuum manifold frame and firmly place the collection plate into the vacuum manifold base. The collection plate fits tightly and evenly into the corner notches of the base Replace the vacuum manifold frame onto the base such that the frame fits evenly on the base. 7

9 ASSAY PROCEDURE 10.3 Place the assay filter plate on top of the vacuum manifold frame such that it is aligned above the collection plate Turn on the vacuum source and press down on the filter plate. Observe the decreasing liquid levels in the filter plate and turn off the vacuum once all filtrate has been collected. 8
