1 The Polyphenol Study Effect of Dietary Polyphenols on Intestinal Microbiota, Intestinal Inflammation and Metabolic Syndrome Principal Investigator: Michael Lefevre, PhD

2 What are Polyphenols? * Polyphenols are a group of chemicals found in many fruits, vegetables, and other plants, such as berries, walnuts, olives, tea leaves, broccoli, grapes and even chocolate! * They are classified as antioxidants, meaning that they remove free radicals from the body. Free radicals are chemicals that can cause damage to cells and tissues in the body.

3 The role of Polyphenols * Many polyphenols have arisen in plants as a defense mechanism against bacterial pathogens and have antibacterial properties * Studies show that consumption of polyphenol- rich foods alter the type of bacteria in the intestines

4 What is the Purpose of the Polyphenol Study? The purpose of this study is to find out if polyphenols affect overall health by modifying the intestinal (gut) bacteria

5 Intestinal (Gut) Bacteria * Changes in our gut bacteria population can have major health consequences, both good and bad

6 Intestinal (Gut) Bacteria Why are they important?? Gut bacteria appear to play a role in our immune systems, metabolism, and insulin sensitivity. Insulin sensitivity is good; insulin resistance is a major risk factor for type 2 diabetes.

7 Insulin Resistance (Sometimes referred to as Metabolic Syndrome) * Is more common in those who are overweight or obese and is associated with high blood pressure, abnormal cholesterol and blood sugar levels. * Insulin resistance is also associated with higher levels of whole body inflammation

8 Diet and Gut Inflammation * The typical Western Diet (high amounts of red meat, fried food, refined grains and foods high in sugar and fat) increases the chance of inflammation in the gut * This inflammation is believed to alter the gut wall in a way that causes it to be more leaky, allowing toxins to cross over the gut wall where they are absorbed into the bloodstream, leading to inflammation throughout the body

9 Diet and Inflammation * This low- grade inflammation is believed to increase the likelihood that a person may develop type 2 diabetes and other medical conditions possibly related to insulin resistance or metabolic syndrome

10 What is the Role of Polyphenols? * Studies in animals show that polyphenols can help protect the gut from inflammation * Do polyphenols affect health by modifying the gut microbiota? Protects the gut from inflammation Polyphenols Alters gut bacteria

11 Participating in The Polyphenol Study * Qualified participants will be provided two different diets. Each diet will be consumed for 6 weeks with a break between the two diets. * All meals, snacks and beverages will be prepared by the Center for Human Nutrition Studies Research Kitchen. * Participants should not eat ANY foods other than what we provide.

12 The Polyphenol Study * This study requires that you eat breakfast and dinner at our beautiful dining facility on the Innovation Campus, northeast of Lee s Market (our address: 650 East 1600 North). * Lunches and weekend meals will be packaged and provided to you in take- out fashion. * This is not a weight- loss study. You will be asked to maintain your weight throughout the study. * The diets are not vegan or vegetarian.

13 DO YOU QUALIFY?? You may be eligible if you: * Are 18 to 70 years of age * Have a body mass index (BMI) in the range of kg/m 2 * Are free of certain chronic diseases * Are willing to eat only the foods that are provided to you by the Center during the two diet periods * Are willing to give up tea, beer, red wine, supplements containing tea extracts, and supplements containing other polyphenols during the study period * Are willing to provide samples for study (blood, urine, stool) Compensation up to $800 upon completion of study
